The hevea planting material nursery is often hindered by suboptimal performance of rootstock. Rootstock nursery always produce discarded seedlings due to attacks by white root disease (WRD) caused Rigidoporus microporus, resulting in short and stunted root morphology. Endophytic fungi from the Dark Septate Endophyte (DSE) filtrate can enhance plant growth and physiological hevea seedling. However, the effect of DSE isolate filtrate from North Sumatra as biostimulant on hevea seedling remain uncertain. This research was conducted to analyze the morphophysiological responses of rubber rootstock application with DSE fungi filtrate.This research was carried out at Plant Protection Laboratory, Experimental Estate of Sungei Putih Research Unit, Indonesian Rubber Research Institute, Galang, Deli Serdang, North Sumatra; and Biology Laboratory of The Republic of Indonesia Defense University, Bogor, West Java, and carried out from July 2023 to May 2024. A Randomized Complete Block Design (RCBD) with four treatments of extracellular filtrate DSE isolate (i.e. control (non DSE), DSE KHPSG isolate, DSE KLAJI isolate, DSE KRPPT isolate) was employed. The results revealed that DSE isolate filtrate application significantly enhanced height, diameter, number of leaves in 3 weeks and plant physiology (proline content, total sugar, root IAA, leaf IAA, total sugar, chlorophyll a, chlorophyll b, and total chlorophyll). The best response in plant growth and physiology is observed with the application of DSE filtrate from KHPSG and KLAJI isolates.
