    Analysis of Caffeine Content in Xanthine in Liberic Type Powder Coffee with Method Ultraviolet Spectrophotometry

    Abstract
    Background: Liberica Coffee is known as a fairly rare type of coffee, as Liberica coffee production is only about 1-2 percent of global coffee production. Coffee contains caffeine, which stimulates the nervous system. But if consumed excessively, the caffeine content in coffee can be detrimental to health. The analysis of caffeine in xanthin can be done using ultraviolet spectroscopic photometry because, in the molecular structure, there are chromophores and auschrome groups. Objective: The objective was to determine if Liberica coffee powder contained caffeine in xanthin and to ascertain whether the analysis of caffeine content in the xanthin powder of Liberica coffee using the ultraviolet spectrophotometry method met validation parameters such as linearity, detection limit, quantitative limit, accuracy, and precision. Method: The research was descriptive, employing purposive sampling. Caffeine levels in xanthin were analyzed using ultraviolet spectroscopic photometry. The method was validated with parameters including precision, accuracy, detection limit, and quantitative limit. Results: The maximum wavelength was 273 nm, and the regression equation was y = 0.0523x + 0.0015 with r = 0.9999. The caffeine levels in samples A, B, C, D, and E were (0.88±0.08)%, (1.03±0.12)%, (1.10±0.033)%, (0.93±0.066)%, and (1.13±0.077)%, respectively. The caffeine levels per sample were 26.4181 mg/trial, 31.0429 mg/trial, 33.1833 mg/trial, 28.1763 mg/trial, and 34.0624 mg/trial. The validation of analytical methods yielded a precision of 1.16%, accuracy of 92.36%, detection limit of 0.1742 μg/mL, and quantitative limit of 0.5809 μg/mL. Conclusion: Liberica coffee powder contains caffeine in xanthin, meeting the standards set by SNI 01-3542-2004 (0.45% to 2% caffeine content) and SNI 01-7152-2006 (150 mg/day to 50 mg/test). The analysis of caffeine levels in xanthin using ultraviolet spectrophotometry met validation parameters including linearity, detection limit, quantitative limit, accuracy, and precision.
