Abstract
This research was conducted to determine the karyotype and ideogram of Lemon fish (Labidochromis caeruleus) and SnowWhite fish (Pseudotropheus socolofi), both of which belong to the same family, which is Cichlidae, and have never been studied their chromosomes before. This research used a descriptive method by the Ideokar 1.3 software and Adobe Photoshop CS6 application. The fish to be tested were purchased from fish sellers in Medan City and then acclimatized for 4 days. Colchicine immersion was performed at 0.005% for 7-8 hours, followed by tissue fixation using a fixative solution for 1 hour and replaced with a carnoy solution for another 1 hour. Then, preparations were made using 50% glacial acetic acid on a hot plate, and Giemsa staining was performed at 2.5%. Data obtained through a microscope were analyzed using IdeoKar 1.3 software and Adobe Photoshop CS6, where the application obtained the number, shape, size of chromosomes, and several other parameters. The results of this study showed that Lemon fish has 26 pairs of metacentric chromosomes. Lemon fish has the largest chromosome size which are 3,4 µm, r-value of 0.852 cM, relative length percentage of 5.370%, F% value of 2.472%, and then the asymmetry index value of HCL 117,3 µm; TF% 45.86%; Ask% 54.13%, and Xci value of 0.45%. Meanwhile, the Snowwhite fish has 24 pairs of chromosomes consisting of 23 pairs of metacentric chromosomes and 1 pair of sub-metacentric chromosomes. The largest chromosome size is 3,9 µm, with an r-value of 0.615%; %RL 6.984%; %F 2.660%, and in the analysis of the asymmetry index, HCL 90,2 µm; TF% 44.56%; Ask% 55.43%, and Xci value of 0.44%.
