    Pengaruh Pemberian Evoo Terhadap Kadar MDA Plasma dan Ekspresi Protein Hsp70, Bcl-2 pada Tikus Putih (Rattus norvegicus) Bunting Model Preeklampsia

    Abstract
    Preeclampsia or hypertension in pregnancy is one of the causes of maternal death, and it tends increasing One of the causes of preeclampsia is an imbalance of antioxidants in the body and can be prevented by giving EVOO, Giving EVOO rich in tocopherol to prevent imbalance between the production of free radicals and antioxidants, especially in pregnant conditions, The design of this research is pre and post test with control goup design in laboratory, Subject of female pregnant white rat, Sprague Dawley strain, BW ± 200g The total of 25 samples consisted of control group (PO) and 4 treattnent groups as preeclampsia model (PI, P2, P3, P4), The preeclamptic model was performed with 6% 3 ml / day NaCI injection on .. to 12th day of the pregnancy period and acute stress of the 18 th day only once, All treattnent groups were given EVOO except PI, from 13th to 1 tjh day, 21 day, all subject were executed Plasma MDA rates decreased significantly after administration of EVOO and significant differences between control and treattnent groups (P value ~ 0,036), Bcl-2 expression increased significantly, especially in the treattnent groups were received EVOO (P value ~ 0,009), The mean TD systole decreased significantly, especially the treattnent groups were given EVOO from before, and the decrease was significantly different with control on day 2dh (P value ~ 0,010), BBW and LB on treattnent groups were given low or moderate dose of EVOO not different from control, 71,43% offetuses died in PI, and 41,94% in P4, Conclusions EVOO able to suppress free radicals in the blood It is recommended to measure other natural antioxidants such as GPx, GSH and vitamin E to determine the role in reducing lipid peroxide after administration of EVOO.
     
    Preeklampsia alau hipertensi dalam kehamilan merupakan salah satu penyebab kernatian ibu, sampai saat ini cenderung meningkat. Salah satu penyebab preeklampsia adalah ketidak seimbangan antioksidan di dalam tubuh dan dapat dicegah dengan pemberian EVOO. Pemberian EVOO yang kaya akan kandungan tokoferol bertujuan untuk mencegah ketidakseimbangan antara produksi radikal bebas dan antioksidan khususnya pada kondisi hamil. Rancangan penelitian ini pre and post test with control goup design di laboratorium. Subyek tikus putih betina bunting, galur Sprague Dawley, BB ±200g. Total sampel 25 ekor terdiri alas kelompok kontrol (PO) dan 4 kelompok perlakuan sebagai model preeklampsia (PI, P2, P3, P4). Model preeklampsia dilakukan dengan injeksi NaCI 6% 3 mllhari pada hari ke 6 - 12 periode kebuntingan dan stres akut hari ke18 sekali saja. Seluruh kelompok perlakuan diberi EVOO kecuali PI, dari hari ke 13 - 19. Hari ke 20, seluruh tikus dieksekusi. Rerata l\1DA plasma menurun secara signifikan setelah pemberian EVOO dan beda nyala antara kelompok kontrol dan perlakuan (P value ~ 0,036). Ekspresi Bcl-2 meningkat cukup kuat secara signifikan, khususnya pada kelompok perlakuan yang mendapat EVOO (P value ~ 0,009). Rerala TD sistole menurun secara signifikan, khususnya kelompok perlakuan yang diberi EVOO dari sebelumnya, dan penurunan terse but beda nyata dengan kontrol pada hari ke 20 (P value ~ 0,010). BBL dan PB kelompok perlakuan yang diberi EVOO dosis rendah maupun sedang tidak beda dengan kontrol, 71,43% fetus mati pada kelompok PI, dan 41,94% pada P4. Kesimpulan EVOO mampu menekan radikal bebas dalam darah. Disarankan untuk mengnkur antioksidan alami lainnya seperti GPx, GSH serta vitamin E untuk mengetahui peranan dalam mereduksi peroksida lemak setelah pemberian EVOO.
