    Deteksi Begomovirus pada Tanaman Cabai (Capsicum annum L.) Berpenyakit di Daerah Tanah Karo Sumatera Utara dengan Teknik PCR

    Abstract
    Begomovirus (from the Geminivirus) is one of the most common virus that causes losses of chili plants in Tanah Karo. The existence of Begomovirus infection effect the chili plant reduce the production, and make a decrease in both the quality and quantity of that. Begomovirus detection only by looking at morphological symptoms could not provide accurate results. It is because Begomovirus has symptoms that are almost identical to other viral diseases. Therefore this research was done to detect Begomovirus molecularly by using PCR technique. A total of 20 leaf samples from chili plant with symptomatic virus were taken from 20 villages in Tanah Karo. Results of the research were morphological symptoms such as vein clearing, thickened and curved leaf bones (cupping), young leaves shrink and bright yellow colored leaves. PCR by using specific primers Begomoviruses indicated the presence of a 1500 Bp DNA fragment in all samples of chili DNA. Anatomical results in the form of stomatal density showed healthy chili plants have greater stomata density compared to Begomovirus infected plants.
     
    Begomovirus (dari kelompok Geminivirus) merupakan salah satu virus penyebab penyakit yang banyak mengakibatkan kerugian pada tanaman cabai yang terdapat di Tanah Karo. Adanya infeksi Begomovirus menyebabkan tanaman cabai tidak dapat melakukan produksi dengan baik sehingga mengakibatkan penurunan baik dari segi kualitas maupun kuantitas tanaman cabai di Tanah Karo. Deteksi Begomovirus hanya dengan melihat gejala morfologinya saja tidak dapat memberikan hasil yang akurat karena Begomovirus memiliki gejala yang hampir sama dengan penyakit akibat virus lainnya. Oleh karena itu penelitian ini dilakukan untuk mendeteksi Begomovirus secara molekuler dengan menggunakan teknik PCR. Sebanyak 20 sampel daun tanaman cabai bergejala virus dikoleksi dari 20 Desa yang ada di Tanah Karo. Hasil penelitian berupa adanya gejala morfologi seperti vein clearing, tulang daun menebal dan melengkung keatas (cupping), daun-daun muda mengecil dan helai daun bewarna kuning cerah. PCR dengan menggunakan primer spesifik Begomovirus menunjukkan adanya fragmen DNA berukuran 1500 Bp pada seluruh sampel DNA tanaman cabai. Hasil anatomi berupa kerapatan stomata menunjukkan tanaman cabai sehat memiliki kerapatan stomata yang lebih besar dibandingkan dengan tanaman terinfeksi Begomovirus.
