dc.description.abstract | Latar belakang: Kanker merupakan penyebab kematian di dunia. Kanker payudara merupakan salah satu jenis kanker terbanyak di Indonesia. Pengobatan kanker payudara biasanya menggunaan obat-obat sintetik yang memiliki efek samping antara lain anemia, alopesia, kardiotoksik dan hepatotoksik. Rimpang teki (Cyperus rotundus Linn.) mengandung banyak senyawa terpenoid yang berpotensi sebagai antikanker. Tujuan penelitian ini untuk mengetahui aktivitas antikanker rimpang teki sehingga dapat dikembangkan untuk pengobatan kanker payudara.
Metode: Pada penelitian ini rimpang teki diekstraksi secara perkolasi menggunakan pelarut etanol, selanjutnya difraksinasi dengan n-heksana, etil asetat dan fraksi air. Aktivitas sitotoksik ditentukan dengan mengamati aktivitas ekstrak ini terhadap sel MCF-7 menggunakan metode MTT assay. Indeks selektivitas (IS) dihitung terhadap sel normal (sel Vero). Siklus sel dan induksi apoptosis dianalisis dengan metode flow cytometry, dan ekspresi protein dilakukan dengan metode imunositokimia.
Hasil: Berdasarkan perlakuan diperoleh hasil uji pada sel MCF-7 aktivitas sitotoksik IC50 ekstrak etanol 122,157 μg/mL, fraksi n-heksana 120,819 μg/mL, fraksi etil asetat 2444,660 μg/mL, fraksi air 2519,264 μg/mL, doksorubisin 462,484 μg/mL dan terhadap sel Vero 221,243 μg/mL. IC50 paling kecil adalah fraksi n-heksana rimpang teki (FnHRT) dengan nilai 120,819 μg/mL dan hasil indeks selektivitas 1,831. Hasil analisis siklus sel menunjukkan bahwa penghambatan terjadi pada fase G0-G1 dan induksi apoptosis terjadi pada apoptosis ahkir (nekrosis awal). Selanjutnya, hasil analisis ekspresi protein menunjukkan bahwa FnHRT mampu menekan ekspresi Cyclin D1, Bcl2 dan meningkatkan ekspresi p53 pada sel MCF-7 bergantung konsentrasi.
Kesimpulan: Berdasarkan hasil yang diperoleh kesimpulan penelitian ini adalah FnHRT mempunyai aktivitas antikanker terhadap sel kanker payudara. Ekstrak ini memiliki mekanisme antikanker melalui penghambatan siklus sel, menginduksi apoptosis, menekan ekspresi protein Cyclin D1, Bcl2 dan meningkatkan ekspresi p53. | en_US |
dc.description.abstract | Introduction: Cancer is one of the causes of morbidity and mortality worldwide. Breast cancer is one of the most common types of cancer in Indonesia. The patients therapy breast cancer followed up with synthetic drug which has side effects such as anemia, alopesia, cardiotoxic and hepatotoxic. Cyperus rotundus Linn. rhizomes contains terpenoid compounds had potential as anticancer. This study aimed to evaluate anticancer activity of Cyperus rotundus Linn, had potential to be developed as anticancer agent. Methode: Cyperus rutundus Linn. rhizomes powder was extracted by percolation using 96% ethanol then fractionated with n-hexane, ethil acetat and water fraction. The cytotoxic activity was determined by observing this extract on MCF-7 cells using MTT assay. Selectivity index (SI) was conducted on normal cells (Vero cells). Cell cycle and apoptosis induction were analyzed by flow cytometry and protein expression was analysed using immunocytochemistry.
Result: Based on studied, IC50 of ethanol extract 122.157 μg/mL, n-hexane fraction 120.819 μg/mL, ethyl acetate fraction 2444.660 μg/mL, water fraction 2519.264 μg/mL, doxorubicin 462.484 μg/mL and Vero cells 221.243 μg/mL. The n-hexane fraction Cyperus rotundus Linn. with a lowest IC50 value was 120.819 μg/mL and selectivity index value 1.831, with accumulation cell cycle in the G0-G1 phase and through induction of apoptosis. Furthermore, the result of an analysis of expression protein Cyclin D1, Bcl2 showed that FnHCR enabled the suppression of Cyclin D1, Bcl2 and improved expression of p53 in MCF-7 cells in a concentration-dependent manner. Conclution: Based on the result study obtained conclutions n-hexane fraction of Cyperus rotundus Linn. rhizome exhibited anticancer activity against MCF-7 breast cancer cells. The mechanisms that underlie anticancer of the extract were due to by inhibition of cell cycle, induction of apoptosis as well as suppression of Cyclin D1, Bcl2 and increase p53 expression. | en_US |
