Pengaruh Pemberian Curcuminoid dalam Mencegah Terjadinya Apoptosis Fibroblas Koklea Rattus Novergicus Model Diabetes Mellitus

Tujuan Membuktikan curcuminoid dapat berperan dalam mencegah dan mengobati kerusakan
fibroblas dinding lateral koklea tikus model Diabetes Melitus (DM) dilihat dari Indeks Apoptosis.
Bahan dan Metode 24 ekor tikus Rattus norvegicus strain Wistar secara acak dibagi dalam
enam kelompok. K1: kelompok kontrol, K2: streptozotocin 12 mg (+), K3: streptozotocin 12 mg
(+), curcuminoid 40 mg/hari selama 5 hari (+), K4: streptozotocin 12 mg (+), curcuminoid 80
mg/hari untuk 5 hari, K5: streptozotocin 12 mg (+), curcuminoid 40 mg/hari selama 10 hari. K6:
Streptozotocin 12 mg (+), curcuminoid 80 mg/hari selama 10 hari. Streptozotocin diberikan
dosis tunggal secara intraperitoneal. Curcuminoid berasal dari Curcuma longa L. (Turmeric),
diberikan secara oral. Seluruh sampel diperiksa dengan metode TUNEL assay untuk melihat
Indeks Apoptosis dalam fibroblas koklea. Data dianalisis secara statistik dengan tingkat
kepercayaan 95%.
Hasil Menunjukkan penurunan Indeks Apoptosis dibandingkan kelompok tanpa curcuminoid
(P<0,05).
Kesimpulan Curcuminoid terbukti efektif dalam mencegah kerusakan pada fibroblas dinding
lateral koklea tikus model DM yang dinilai dari penurunan Indeks Apoptosis. Objective To prove that curcuminoid can play a role in preventing and treating damage to the
cochlear lateral wall fibroblasts in the model of Diabetes Melitus (DM) seen from the Apoptosis
Index.
Materials and Methods 24 Wistar strains of Rattus norvegicus rats were randomly divided into
six groups. K1: control group, K2: streptozotocin 12 mg (+), K3: streptozotocin 12 mg (+),
curcuminoid 40 mg / day for 5 days (+), K4: streptozotocin 12 mg (+), curcuminoid 80 mg / day
for 5 days, K5: streptozotocin 12 mg (+), curcuminoid 40 mg / day for 10 days. K6:
Streptozotocin 12 mg (+), curcuminoid 80 mg / day for 10 days. Streptozotocin is given a single
dose intraperitoneally. Curcuminoid comes from Curcuma longa L. (Turmeric), given orally. All
samples were examined by the TUNEL assay method to see the Apoptosis Index in cochlear
fibroblasts. Data were analyzed statistically with a 95% confidence level.
Results showed a decrease in the Apoptosis Index compared to the group without curcuminoid
(P <0.05).
Conclusion Curcuminoid has been shown to be effective in preventing damage to the cochlear
lateral wall fibroblasts in DM models that are assessed by a decrease in the Apoptosis Index.
