    Aplikasi Kolkisin Terhadap Variasi Morfologi dan Fisiologi Tanaman Bawang Putih (Allium sativum L.) Doulu

    Abstract
    Productivity of Doulu’s garlic is low. It is caused by small size of bulbs, Doulu’s garlic is rarely found in the market, as due to high cost productivity. Therefore the farmer is not interested to grow garlic. An effort that can be done by using the plant mutagen, colchicine. This study aims to determine the morphological and physiological variation of Doulu’s garlic plants after colchicine application. The design experimental was a Factorial Group Random Design with 20 combinations and 3 replications. The first factor was concentration of colchicine (D): 0% (D0), 0.1% (D1), 0.2% (D2), and 0.3% (D3). The second factor was soaking time (T): 0 (T0), 6 (T1), 12 (T2), 18 (T3), and 24 (T4) hours. The results showed the highest height of plant, number of leaves, and dry weight of garlic bulbs were produced by plant in the treatment without extending colchicine with soaking for 12 hours (D0T2). The best number of roots and fresh weight of garlic bulbs were produced in the application of 0.3% colchicine without immersion (D3T0). The highest root length and stomata density were produced by plant in the treatment without extending colchicine with soaking for 18 hours (D0T3). This is different from the total chlorophyll content of garlic plants which has increased after being applied with 0.1% colchicine for 24 hours (D1T4) of 22.26 mg/l compared to the control group. The number of normal chromosomes Doulu’s garlic was 2n = 16.
     
    Produktifitas bawang putih Doulu masih rendah. Hal ini disebabkan ukuran umbi kecil, bawang putih Doulu jarang ditemukan di pasar, serta biaya produktifitas tinggi. Oleh karena itu, petani kurang berminat untuk menanam bawang putih. Upaya yang dapat dilakukan ialah memutasikan tanaman menggunakan kolkisin. Penelitian ini bertujuan untuk mengetahui variasi morfologi dan fisiologi tanaman bawang putih Doulu terhadap aplikasi kolkisin. Rancangan penelitian yang digunakan adalah Rancangan Acak Kelompok Faktorial dengan 20 kombinasi dan 3 ulangan. Faktor pertama adalah konsentrasi kolkisin (D): 0% (D0), 0,1% (D1), 0,2% (D2), dan 0,3% (D3). Faktor kedua adalah waktu perendaman (T): 0 (T0), 6 (T1), 12 (T2), 18 (T3), dan 24 (T4) jam. Hasil penelitian menunjukkan tinggi tanaman, jumlah daun, dan berat kering umbi bawang putih yang tertinggi dihasilkan pada perlakuan tanpa pemberian kolkisin dengan perendaman selama 12 jam (D0T2). Jumlah akar dan berat basah umbi terbaik dihasilkan pada perlakuan kolkisin 0,3% tanpa perendaman (D3T0). Panjang akar dan kerapatan stomata tertinggi dihasilkan pada perlakuan tanpa pemberian kolkisin dengan perendaman selama 18 jam (D0T3). Hal ini berbeda dengan kandungan klorofil total tanaman bawang putih yang mengalami peningkatan setelah diaplikasikan dengan kolkisin 0,1% selama 24 jam (D1T4) sebesar 22, 26 mg/l dibandingkan dengan kelompok kontrol. Jumlah kromosom normal bawang putih Doulu adalah 2n = 16.
