    Pengaruh Pemberian Curcuminoid Terhadap Ekspresi Transforming Growth Factor Beta (TGF-β) pada Fibroblas Koklea Rattus Norvegicus Model Diabetes Melitus

    Abstract
    Pendahuluan: TGF-β berperan dalam proses inflamasi dan peningkatan radikal bebas melalui jalur PKC dalam keadaan hiperglikemia yang mengakibatkan komplikasi antara lain mikroangiopati yang diketahui terjadi terutama pada organ-organ dengan mikrovaskularisasi termasuk koklea. Curcumin sebagai antiinflamasi dan antioksidan telah terbukti aman digunakan pada banyak penyakit, termasuk diabetes, diharapkan mampu menurunkan ekspresi TGF-β yang diduga meningkat pada dinding lateral koklea tikus model diabetes melitus. Tujuan: Penelitian ini bertujuan untuk melihat peran curcumin dalam menurunkan ekspresi TGF- β pada dinding lateral koklea tikus model diabetes mellitus Metode: 6 kelompok tikus dengan jumlah sampel yang sama (masing-masing 4 ekor), kelompok 1 adalah kontrol negatif, kelompok 2 adalah kontrol positif (tikus yang diinjeksi Streptozotocin), kelompok 3 dan 4 secara berurutan adalah tikus model diabetes yang diberikan curcumin 200 mg/kgBB/hari (dosis kecil) dan curcumin 400 mg/kgBB/hari (dosis besar) selama 5 hari, kelompok 5 dan 6 secara berurutan adalah tikus model diabetes yang diberikan curcumin dosis kecil dan curcumin dosis besar selama 10 hari. Seluruh sampel lalu diterminasi dan di nekropsi untuk pengambilan jaringan kokleanya, dan kemudian diperiksa ekspresi TGF- β dengan pemeriksaan imunohistokimia. Hasil: Dijumpai perbedaan ekspresi TGF- β yang bermakna (p<0.05)pada tikus model diabetes (kelompok 2) dibandingkan kontrol (kelompok 1), serta bila kelompok 2 dibandingkan dengan kelompok 5 dan dengan keompok 6. Tidak ada perbedaan bermakna ekspresi TGF-β (p>0.05) pada kelompok 3 dibandingkan dengan kelompok 5, namun terdapat perbedaan bermakna ekspresi TGF-β (p<0.05) kelompok 4 dibandingkan dengan kelompok 6.
     
    Introduction: TGF-β plays a role in the inflammatory process and increases free radicals through the PKC pathway in a hyperglycaemic state which results in complications such as microangiopathy known to occur mainly in organs with microvascularization including cochlea. Curcumin as an anti-inflammatory and antioxidant has been shown to be safe to use in many diseases, including diabetes, is expected to decrease the expression of TGF-β, which suspected to be elevated in the lateral wall of the cochlea diabetic model rats. Objective: This study aims to examine the role of curcumin in decreasing TGF-β expression in the lateral wall of the rat cochlea model of diabetes mellitus. Methods: 6 groups of rats with the same number of samples (4 each), group 1 was negative control, group 2 was positive control (Streptozotocin injected rats), group 3 and 4 were diabetic model rats given curcumin 200 mg / kgBW / day (small dose) and curcumin 400 mg / kgBW / day (large dose) for 5 days, group 5 and 6 respectively were diabetic model rats given small doses of curcumin and large doses of curcumin for 10 days. All samples were then terminated and necropsed for the removal of their cochlear tissue, and then examined the TGF-β expression by immunohistochemical examination. Results: There was a significant difference in TGF-β expression (p <0.05) in diabetic model rats (group 2) compared to control (group 1), and the same result (p<0.05) when group 2 was compared with group 5 and with group 6. There was no significant difference in TGF- β (p> 0.05) in group 3 compared to group 5, but there was a significant difference in TGF-β expression (p <0.05) group 4 compared to group 6.
