    Inisiasi dan Pertumbuhan Kalus pada Kultur Daun Apel (Pyrus malus L.) dalam Media MS dengan Kombinasi BAP dan NAA

    Abstract
    The research was to determine the effect of BAP and NAA in initiating callus of the leaf explants of apple (Pyrus malus L.) cv. Manalagi. from the leaf explants of apple. This research was conducted from April to December 2016 at Laboratory of Plant Physiology and Tissue Culture, Department of Biology, Faculty of Mathematics and Natural Sciences, University of North Sumatra, Medan. The research method is using a completely randomized factorial design with two factors namely the variation combination of BAP (0; 0.1; 0.2; 0.3 mg / L) and a variation of the combination of NAA (0; 0.2; 0.4; 0 , 6 mg / L) in Murashige and Skoog medium. The results showed that the percentage of callus cultures were 87.5%, with two types of callus that was compact textures 57.14% and friable ones 42.86%, while colors visualizing was dominated by brownish white 52,4%, white 30,95% and white yellowish 16,67%. The best of time of initiation and fresh weight on B3N2 treatment (0.3 BAP and NAA 0.4 mg / L), while the highest of the dry weight of callus was on B3N3 treatment (0.3 BAP and NAA 0.6 mg / L).
     
    Penelitian ini untuk mengetahui pengaruh BAP dan NAA dalam menginisiasi kalus dari ekplan daun apel (Pyrus malus L.) cv. Manalagi. Penelitian dilakukan mulai April sampai dengan Desember 2016 di Laboratorium Fisiologi dan Kultur Jaringan Tumbuhan, Departemen Biologi, Fakultas Matematika dan Ilmu Pengetahuan Alam, Universitas Sumatera Utara, Medan. Metode penelitian dilakukan menggunakan rancangan acak lengkap faktorial dengan dua faktor, yaitu variasi kombinasi BAP (0; 0,1; 0,2; 0,3 mg/L) dan variasi kombinasi NAA (0; 0,2; 0,4; 0,6 mg/L) dalam media Murashige dan Skoog. Hasil penelitian menunjukkan bahwa persentase kultur yang membentuk kalus sebesar 87,5%, dengan dua tipe kalus yaitu kalus bertekstur kompak 57,14% dan kalus bertekstur remah 42,86%, sedangkan visualisasi warna kalus didominasi oleh warna putih kecoklatan 52,4%, putih 30,95%, dan warna putih kekuningan 16,67%. Waktu inisiasi dan berat basah kalus yang terbaik adalah pada perlakuan B3N2 (BAP 0,3 dan NAA 0,4 mg/L), sedangkan berat kering kalus yang tertinggi pada perlakuan B3N3 (BAP 0,3 dan NAA 0,6 mg/L).
