Penetapan Kadar Kotrimoksazol dengan Metode Kromatografi Cair Kinerja Tinggi (KCKT)

Abstract

Kotrimoksazol is combination mixture sulfametoksazol and trimethoprim between 5:1 to bacterisid with the spectrum bigger than sulfonamide.Trimetoprim and sulfametoksazol will survive enzimatik obligat reaction with 2 steps so this combination will give sinergistik effect. The qualitatif test of each drug was done to suspension sulfametoksazol and trimetoprim solution to system HPLC with coloum, mobile phase, flow rate and detector same as included in United States Pharmacopeia (USP) 34TH Edition 2011. The requirement oral suspension of United States Pharmacopeia (USP) 34TH Edition namely containing sulfametoxazole and trimetoprim not less than 90.0 % and not more than 110.0 % from the label declared. The validation test of the tablet mixture who made by researcher showed the concentration 101.17% for sulfametoxazole and for trimetoprim showed the concentration 105.14%.

Kotrimoksazol merupakan kombinasi dari sulfametoksazol dan trimetoprim dengan perbandingan 5:1, bersifat bakterisid dengan spektrum kerja lebih lebar dibandingkan dengan sulfonamida. Trimetoprim dan sulfametoksazol menghambat reaksi enzimatik obligat pada dua tahap yang berurutan pada bakteri sehingga kombinasi kedua obat memberikan efek sinergistik. Pengujian ini bertujuan untuk menetapkan kadar sulfametoksazol dan trimetoprim dalam sediaan suspensi menggunakan kromatografi cair kinerja tinggi (KCKT) dengan kolom, fase gerak, laju alir dan detektor yang tertera pada United States Pharmacopeia (USP) Edisi XXXIV tahun 2011. Persyaratan kadar suspensi kotrimoksazol menurut USP Edisi XXXIV tahun 2011 baik untuk sulfametoksazol maupun trimetoprim mengandung tidak kurang dari 90,0% dan tidak lebih dari 110,0% dari jumlah yang tertera pada etiket. Hasil pengujian menunjukkan bahwa kadar suspensi kotrimoksazol yang diuji tersebut memenuhi persyaratan yaitu untuk kadar sulfametoksazol sebesar 101,17% dan kadar trimetropim sebesar 105,14%.
