    Uji Aktivitas Antikanker Ekstrak Daun Afrika (Vernonia amygdalina Del.) Terhadap Model Sel Kanker Kolon WiDr

    Abstract
    Kanker kolon merupakan kanker dengan jumlah terbanyak ketiga pada pria dan kedua pada wanita. Pengobatan kanker di antaranya menggunakan kemoterapi, namun sejumlah obat kemoterapi telah mengalami resisten. Daun Afrika (Vernonia amygdalina Del.) merupakan salah satu tumbuhan yang banyak dilaporkan dapat menghambat pertumbuhan berbagai model sel kanker. Penelitian ini bertujuan untuk menguji aktivitas antikanker ekstrak aktif Daun Afrika (Vernonia amygdalina Del.) terhadap model sel kanker kolon WiDr. Penelitian ini merupakan penelitian eksperimental in vitro dengan menggunakan desain posttest only control group. Subjek penelitian merupakan model sel kanker kolon WiDr. Metode penelitian dimulai dari karakterisasi dan skrining fitokimia simplisia Daun Afrika (Vernonia amygdalina Del.), pembuatan ekstrak secara maserasi bertingkat menggunakan pelarut n-heksana, etil asetat, dan etanol, skrining fitokimia ekstrak, uji sitotoksik terhadap sel WiDr dengan metode MTT (Microculture Tetrazolium Technique), uji indeks selektivitas terhadap sel WiDr dan Vero, uji penghambatan siklus sel dan apoptosis dengan metode flowsitometri, serta uji ekspresi COX-2 dan cleaved caspase-3 dengan metode imunositokimia. Hasil uji sitotoksik (IC50) ENDA, EEADA, dan EEDA terhadap sel WiDr berturut-turut sebesar > 500 μg/mL, 9,086 ± 0,431 μg/mL, dan 321,131 ± 9,902 μg/mL, EEADA tidak selektif terhadap sel WiDr (IS=1,47), penghambatan siklus sel WiDr pada fase G2-M, pemacuan apoptosis sel WiDr, penekanan ekspresi COX-2 (p<0,05), dan peningkatan ekspresi cleaved caspase-3 (p<0,05). EEADA memiliki aktivitas antikanker sangat kuat terhadap sel WiDr, melalui penghambatan siklus sel pada fase G2-M, pemacuan apoptosis, penekanan ekspresi COX-2, dan peningkatan ekspresi cleaved caspase-3.
     
    Colon cancer is the third most common cancer in men and second in women. Cancer treatments include chemotherapy, but some chemotheraphy drugs are resistant. African Leaf (Vernonia amygdalina Del.) is one of the most widely reported plant that can inhibit the growth of various cancer cell lines. This study aims to examine the anticancer activity of African Leaf (Vernonia amygdalina Del.) active extract against WiDr colon cancer cell line. This research is an experimental research in vitro using posttest only control group design. Research subjects are WiDr colon cancer cell line. The research method starts from the characterization and phytochemical analysis of African Leaf (Vernonia amygdalina Del.) simplicia, maceration of the extract using n-hexane, ethyl acetate, and ethanol, phytochemical analysis of extracts, cytotoxicity assay against WiDr cell using MTT method (Microculture Tetrazolium Technique), selectivity index assay of WiDr and Vero cells, cell cycle assay and apoptosis assay using flowcytometry methods, and expression of COX- 2 and cleaved caspase-3 using immunocytochemistry method. The cytotoxicity assay (IC50) ENDA, EEADA, and EEDA against WiDr cell respectively are > 500 μg/mL, 9.086 ± 0.431 μg/mL, and 321.131 ± 9.902 μg/mL, EEADA not selective to WiDr cell (SI = 1 , 47), WiDr cell cycle arrest at G2-M phase, pacing WiDr cell apoptosis, decrease of COX-2 expression (p <0.05), and increased of cleaved caspase-3 expression (p <0.05). EEADA has a very strong anticancer activity against WiDr cell, through cell cycle arrest at G2-M phase, pacing apoptosis, decrease of COX-2 expression, and increased of cleaved caspase-3 expression.
