dc.description.abstractPendahuluan Penelitian ini bertujuan untuk menunjukkan potensi kurkumin sebagai antioksidan dengan meningkatkan ekspresi Superoxide Dismutase (SOD) di dinding lateral koklea pada model tikus ototoksik. Bahan dan Metode Sebanyak 24 Rattus norvegicus dibagi menjadi 8 kelompok: kelompok 1 (kelompok kontrol), kelompok 2 (gentamisin (+)), kelompok 3 (gentamicin + curcumin 20 mg / hari), kelompok 4 (gentamisin + curcumin 40 mg / hari ) kelompok 5 (gentamicin + curcumin 20 mg / hari selama 7 hari), kelompok 6 (gentamicin + curcumin 40 mg / hari selama 7 hari), kelompok 7 (kurkumin 20 mg / hari selama 3 hari + gentamisin), dan kelompok 8 (kurkumin 40 mg / hari selama 3 hari + gentamisin). Setelah pembagian, tikus diakhiri untuk mengukur indeks apoptosis dengan uji imunohistokimia di fibroblas dinding lateral koklea. Data dianalisis dengan menggunakan ANOVA dan p <0,05 digunakan sebagai cut-off untuk signifikansi statistik. Hasil Pemberian gentamisin menunjukkan penurunan ekspresi SOD yang signifikan (p <0,05). Pemberian kurkumin dalam dosis tinggi (40 mg) dan kelompok pencegahan menunjukkan perbedaan yang signifikan dibandingkan kelompok tanpa pemberian kurkumin. Kesimpulan Penelitian ini menyimpulkan bahwa kurkumin mampu meningkatkan ekspresi SOD dan memiliki manfaat preventif terhadap model tikus ototoksik. Pemberian kurkumin bergantung pada dosis dan durasinya.en_US
dc.description.abstractIntroduction This study aimed to show the potency of curcumin as an antioxidant that increase the expression of Superoxide Dismutase (SOD) in cochlea lateral wall in ototoxic rat models. Materials and Methods A total of 24 Rattus norvegicus were divided into 8 groups: group 1 (control group), group 2 (gentamicin (+)), group 3 (gentamicin + curcumin 20 mg/day), group 4 (gentamicin + curcumin 40 mg/day), group 5 (gentamicin + curcumin 20 mg/day for 7 days), group 6 (gentamicin + curcumin 40 mg/day for 7 days), group 7 (curcumin 20 mg/day for 3 days + gentamicin), and group 8 (curcumin 40 mg/day for 3 days + gentamicin). After the division, the rats were terminated in order to measure the apoptotic index with TUNEL assay in the fibroblasts of the cochlea lateral walls. The Data were analysed by using ANOVA and p < 0.05 was used as the cut-off for statistical significance. Results Administration of gentamicin decreased the SOD expression significanty (p < 0.05). Treatment of curcumin in higher dose (40 mg) and prevention group showed significant differences compared to group without curcumin treatment Conclusion This study concluded that curcumin is able to increase SOD expression and has a preventive benefit toward the ototoxic rat models. The administration of curcumin depended on the dose and duration.en_US
