    Efek Latihan Fisik Terhadap Ekspresi Gen Pgc1a dan Tingkat Stres Oksidatif pada Hewan Model Penuaan Yang Diinduksi D-Galaktosa

    Abstract
    Oxidative stress is caused by an imbalance of free radicals to the level of antioxidants in the body. These conditions cause lipid peroxidation and products produced include malondialdehyde (MDA). D-galactose is a substance that causes oxidative stress that occurs in the subcellular, especially in the mitochondria. Mitochondrial biogenesis is influenced by Pgc1a which is a coactivator of the transcription process of several genes that play a central role in mitochondrial regulation. Physical exercise can induce molecular pathways to increase Pgc1a expression, then induce protein expression in the process of mitochondrial biogenesis and decrease in oxygen reactive species (ROS). This study aims to observe the effects of physical exercise on Pgc1a gene expression from the hippocampus tissue using Real Time-PCR and oxidative stress levels by measuring MDA in serum D-galactose-induced mice. Fifteen male mice were divided into 3 groups. Group 1 was injected imitaperitonially with aquabides, group 2 was injected with D-galactose 150 mg/kgBW, group 3 was given Dgalactose 150 mg/kgBW and physical exercise (running wheel). There was no significant difference in MDA levels in groups 1 and 2 (p=0.95). There was a significant difference in MDA levels (p 0.01) in groups 2 and 3. There was no significant difference (p = 0.29) on Pgc1a gene expression in groups 2 and 3.
     
    Stres oksidatif disebabkan oleh ketidakseimbangan radikal bebas terhadap tingkat antioksidan dalam tubuh. Kondisi tersebut menyebabkan timbulnya peroksidasi lipid dan produk yang dihasilkan antara lain malondialdehid (MDA). D-galaktosa adalah satu bahan yang menimbulkan stres oksidatif yang terjadi di subseluler terutama di mitokondria. Biogenesis mitokondria dipengaruhi oleh Pgc1a yang merupakan co-aktivator proses transkripsi beberapa gen yang memainkan peran sentral dalam regulasi mitokondria. Latihan fisik dapat menginduksi jaras molekuler terhadap peningkatan ekspresi Pgc1a, lalu menginduksi ekspresi protein pada proses biogenesis mitokondria dan penurunan species oxygen reactive (ROS). Penelitian ini bertujuan untuk mengamati efek latihan fisik terhadap ekspresi gen Pgc1a dari jaringan hipokampus dengan menggunakan Real Time-PCR dan tingkat stres oksidatif dengan cara mengukur MDA pada serum mencit yang diinduksi D-galaktosa. Lima belas mencit jantan dibagi menjadi 3 kelompok. Kelompok 1 diinjeksi secara imtraperitonial dengan aquabides, kelompok 2 diinjeksi D-galaktosa 150 mg/kgBB, kelompok 3 diberi D-galaktosa 150 mg/kgBB dan latihan fisik (running wheel). Tidak ada perbedaan signifikan kadar MDA kelompok 1 dan 2 p=0,95. Ada perbedaan signifikan kadar MDA (p=0,01) kelompok 2 dan 3. Tidak ada perbedaan yang signifikan (p=0,29) terhadap ekspresi gen Pgc1a pada kelompok 2 dan 3.
