    Uji Aktivitas Hipoglikemik Minuman Gambir dengan Berbagai Metode Ekstraksi Gambir (Uncaria gambir Robx.) pada Mencit yang Diinduksi Aloksan

    Abstract
    Diabetes is a serious chronic disease characterized by hyperglycemic conditions and can causes complications in various parts of the body and increase the risk of early death. Gambier is one of Indonesia's superior commodities which known have bioactive compounds and hypoglycemic effects and can be used as raw material in making gambier drinks. The purpose of this study was to find out the best extraction method of gambier to produce the best gambier drinks and to determine the hypoglycemic activity of gambier drinks and the best dose to decrease blood glucose levels and to improve the pancreatic conditions of alloxan-induced mice. In this study, gambier were extracted by 3 types of solvents ie. distiled water, ethyl acetate, and ethanol. The best gambier drinks then were tested on 5 treatment groups wich consisting of 5 healthy male mice. Group 1 was given 0,5% CMC with dose 1%/bw, group 2 was given metformin 65 mg/kg bw, groups 3, 4 and 5 were given gambier drinks with dosage 100, 200, and 300 mg/kg bw respectively. The results showed that extraction using distiled water produced the best gambier drinks. Gambier drinks with dosage 100, 200 and 300 mg/kg bb given for 15 days decreased the blood glucose level 27.69%, 38.75%, and 50.62% respectively, and increased the body weight 13,14%, 10, 91%, and 10,18% respectively. Treatment with gambier drinks was proved able to improve the condition of pancreatic langerhans island of alloxan-induced mice and indicated that gambier drinks have hypoglycemic activity.
     
    Diabetes merupakan penyakit kronis serius yang ditandai dengan kondisi hiperglikemik dan dapat menyebabkan komplikasi pada berbagai bagian tubuh serta meningkatkan risiko kematian secara dini. Gambir merupakan salah satu komoditi unggulan Indonesia yang diketahui mengandung senyawa bioaktif yang memiliki efek hipoglikemik dan dapat digunakan sebagai bahan baku pembuatan minuman gambir. Tujuan penelitian ini adalah untuk mengetahui metode ekstraksi gambir yang menghasilkan minuman gambir terbaik, mengetahui aktivitas hipoglikemik minuman gambir dan dosis terbaik untuk menurunkan kadar glukosa darah dan memperbaiki kondisi pankreas mencit yang diinduksi aloksan. Gambir diekstraksi dengan 3 jenis pelarut yaitu akuades, etil asetat, dan etanol. Minuman gambir terbaik diujikan secara in vivo pada 5 kelompok perlakuan yang masing-masing terdiri atas 5 ekor mencit jantan. Kelompok 1 diberikan larutan CMC 0,5% sebanyak 1% per bb, kelompok 2 metformin 65 mg/kg bb, kelompok 3, 4 dan 5 diberi minuman gambir 100, 200, dan 300 mg/kg bb. Hasil penelitian menunjukkan bahwa ekstraksi gambir dengan akuades menghasilkan minuman gambir terbaik. Pemberian minuman gambir dosis 100, 200 dan 300 mg/kg bb selama 15 hari pada mencit diabetes menurunkan kadar glukosa darah masing-masing 27,69%, 38,75%, dan 50,62%, serta meningkatkan berat badan 13,14%, 10, 91%, dan 10,18%. Pemberian minuman gambir terbukti mampu memperbaiki kondisi pulau langerhans pankreas mencit dan mengindikasikan bahwa minuman gambir memiliki aktivitas hipoglikemik.
