    Induksi Tunas Mikro Asam Gelugur (Garcinia atroviridis Griff): Pengaruh Jenis Eksplan Dan Zat Pengatur Tumbuh

    Abstract
    The study aims to determine the effect of different types of explants and combination of NAA and BAP growth regulators and their interaction between them on the fornation of micro shoots of the asam gelugur plant (Garcinia Atroviridis Griff). This research was carried out in the tissue culture laboratory of the Faculty of Agriculture, Universitas Sumatera Utara, Indonesia, starting in February 2018 until June 2018. The design used was a completely randomized design with 2 treatment factors, namely a combination of NAA and BAP doses in the media with 9 levels, namely G1 0.2 ml /L NAA + 0.5 ml /L BAP; G2 0.2 ml /L NAA + 1 ml /L BAP; G3 0.2 ml /L NAA + 1.5 ml /L BAP; G4 0.4 ml /L NAA + 0.5 ml /L BAP; G5 0.4 ml /L NAA + 1 ml /L BAP; G6 0.4 ml /L NAA + 1.5 ml /L BAP; G7 0.6 ml /L NAA + 0.5 ml /L BAP; G8 0.6 ml /L NAA + 1 ml /L BAP; G9 0.6 ml /L NAA + 1.5 ml /L BAP while the second factor was the type of explant shoot and node, with 3 replications. The results of the research, showed that the addition of 0.2 ml/L NAA + 1 ml/L BAP in MS media had a significant effect on the age of appearance, number of shoots, length of shoots, and number of leaves. The best type of explant was the nodus, while the best interaction for both, shown by the best response to the length of shoots and the number of explants was the application of 2 ml/L NAA + 1 ml/L BAP to the nodus explant.
     
    Penelitian ini bertujuan untuk mengetahui pengaruh perbedaan jenis eksplan dan kombinasi dosis zat pengatur tumbuh NAA dan BAP beserta interaksi antara keduanya terhadap pembentukan tunas mikro tanaman asam gelugur (Garcinia Atroviridis Griff). Penelitian ini dilaksanakan di Laboratorium Kultur Jaringan Fakultas Pertanian Universitas Sumatera Utara Indonesia. Dimulai pada bulan Februari 2018 sampai dengan Juni 2018. Rancangan yang digunakan adalah rancangan acak lengkap dengan 2 faktor perlakuan yaitu kombinasi dosis NAA dan BAP dalam media dengan 9 taraf yaitu G1 0,2 mg/l NAA + 0,5 mg/l BAP; G2 0,2 mg/l NAA + 1mg/l BAP; G3 0,2 mg/l NAA + 1,5 mg/l BAP; G4 0,4mg/l NAA + 0,5 mg/l BAP; G5 0,4 mg/l NAA + 1mg/l BAP; G6 0,4 mg/l NAA + 1,5 mg/l BAP; G7 0,6mg/l NAA + 0,5 mg/l BAP; G8 0,6 mg/l NAA + 1mg/l BAP; G9 0,6 mg/l NAA + 1,5 mg/l BAP sedangkan faktor ke dua yaitu jenis eksplan pucuk dan nodus dengan 3 ulangan. Hasil penelitian menunjukkan dengan perlakuanpenambahan 0,2 mg/l NAA + 1mg/l BAP dalam media MS memberikan pengaruh nyata terhadap umur muncul tunas, jumlah tunas, panjang tunas, dan jumlah daun. Untuk jenis eksplan, hasil yang terbaik terdapat pada ekplan nodus, sedangkan interaksi untuk keduanya menunjukkan respon terbaik terhadap panjang tunas dan jumlah eksplan dengan menggunakan eksplan nodusdan pemberian ZPT 2 mg/l NAA + 1mg/l BAP.
