    Pengaruh Pemberian Curcuminoid Terhadap Ekspresi NFkB pada Fibroblas Koklea Rattus Novergicus Model Diabetes Mellitus

    Abstract
    Objective: This study aims to prove curcuminoid as an effective and safe fitofarmaka to prevent damage on cochlear fibroblasts due to diabetes mellitus. Methods: Rats were divided randomly into six groups, consisting of a negative control group, and treatment groups that was devided into a diabetes induced group only (by using streptozotocin) and the other treatment groups that was given curcuminoid after induction of diabetes mellitus. All rats were terminated then the cochlear tissue was taken, after that we did the immunohistochemical preparations to assess NFkB expression in cochlear fibroblasts. Results: There is an increase in NFκB expression on cochlear fibroblasts of diabetes induced group only without administration of curcuminoids. The administration of curcuminoid 80 mg for 8 days showed a statistically significant decrease in NFκB expression (p <0.05) compared to other treatment groups. Conclusion: Curcuminoid can reduce NFκB expression in cochlear fibroblasts of diabetes mellitus rats.
     
    Tujuan: Penelitian ini bertujuan untuk membuktikan curcuminoid sebagai fitofarmaka yang efektif dan aman untuk mencegah kerusakan fibroblas koklea akibat diabetes mellitus. Metode: Tikus jantan dibagi secara acak menjadi enam kelompok, yang terdiri dari kelompok kontrol negatif, dan kelompok perlakuan yang hanya diinduksi diabetes mellitus dengan streptozotocin serta kelompok perlakuan yang diberi curcuminoid setelah induksi diabetes melitus. Semua tikus yang diterminasi kemudian dilakukan pengambilan jaringan koklea lalu dibuat sediaan dan pengecatan imunohistokimia untuk dilakukan penilaian ekspresi NFkB pada fibroblas koklea. Hasil: Terjadi peningkatan ekspresi NFkB pada fibroblas koklea yang hanya diinduksi diabetes mellitus tanpa pemberian curcuminoid. Pemberian curcuminoid 80 mg selama 8 hari menunjukkan penurunan ekspresi NFkB yang bermakna secara statistik (p<0.05) dibandingkan kelompok perlakuan lainnya. Kesimpulan: Curcuminoid dapat menurunkan ekspresi NFkB pada fibroblas koklea tikus model diabetes mellitus.
