    Deteksi Virus Potato Virus Y (PVY) pada Tanaman Kentang di Tanah Karo, Sumatera Utara Menggunakan Teknik Molekuler

    Abstract
    Potato virus Y (Potyviridae; Potyvirus) is one of the viruses that attack the potato plant. This virus attack can cause significant losses, the loss is a decrease in yield of potato tubers in quality and quantity. Diagnosis of PVY virus based on symptoms is difficult to determine the identity of a virus. Therefore, this study aims to detect the PVY virus in potato plants with molecular techniques. A total of 20 leaf samples were randomly collected from symptomatic potato crops at each site. Parameters observed were symptomatic, symptomatic plant stomatal density and PVY detection with RT-PCR. The results showed that the symptoms found in the field varied in potato leaf, such as mosaic, leaf distortion, rugose, vein clearing and leaf malformation. RT-PCR using PVY primers succeeded in amplifying PVY DNA taken in 20 villages in Tanah Karo showing a fragment of 801pb and in healthy potato plants having greater stomatal density than symptomatic plants. The results showed that the identification of the virus should be considered as an important limitation of potato production in Tanah Karo.
     
    Potato virus Y (Potyviridae; Potyvirus) merupakan salah satu virus yang menyerang tanaman kentang. Serangan virus ini dapat menimbulkan kerugian yang berarti, kerugian tersebut berupa penurunan hasil panen umbi kentang secara kualitas maupun kuantitas. Diagnosis virus PVY berdasarkan gejala sulit untuk menentukan identitas suatu virus tersebut. Oleh karena itu penelitian ini bertujuan mendeteksi virus PVY pada tanaman kentang dengan teknik molekuler. Sebanyak 20 sampel daun dikoleksi secara acak dari tanaman kentang yang bergejala di setiap lokasi. Parameter yang di amati adalah gejala, kerapatan stomata tanaman yang bergejala dan deteksi PVY dengan RT-PCR. Hasil penelitian menunjukkan bahwa gejala yang ditemukan di lapangan bervariasi pada daun kentang, seperti mosaik, distorsi daun, rugose, vein clearing dan malformasi daun. RT-PCR menggunakan primer PVY berhasil mengamplifikasi DNA PVY yang diambil pada 20 desa di Tanah Karo dengan menunjukkan adanya fragmen pada ukuran 801pb dan pada tanaman kentang sehat memiliki kerapatan stomata yang lebih besar dibanding tanaman yang bergejala. Hasil penelitian menunjukkan bahwa teridentifikasinya virus tersebut harus dipertimbangkan sebagai pembatas penting produksi kentang di Tanah Karo.
