    Isolasi dan Penentuan Struktur Senyawa Kimia Fraksi Etil Asetat dari Daun Durian (Durio zibethinus L.) yang Mempunyai Aktivitas Antidiabetes

    Abstract
    Penelitian mengenai daun durian Durio zibethinus L. telah dilakukan. Penelitian ini bertujuan untuk mengetahui struktur senyawa kimia yang terdapat pada daun Durio zibethinus L. dan yang mempunyai aktivitas antidiabetes dengan metode penghambatan enzim α-glukosidase. Penelitian ini dimulai dengan melakukan ekstraksi daun durian dengan pelarut etanol 96%. Ekstrak kental etanol dilakukan partisi dengan pelarut n-heksan, etil asetat dan air. Selanjutnya masing-masing fraksi dilakukan skrining fitokimia, dan pengujian antidiabetes. Hasil pengujian antidiabetes paling tinggi diperoleh pada fraksi etil asetat. Selanjutnya fraksi etil asetat dilakukan isolasi, pemurnian, dan dikarakterisasi dengan metode spektrofotometri UV, IR, NMR (1D dan 2D), dan LC-MS. Fraksi etil asetat dipisahkan dengan kromatografi kolom, eluen yang digunakan adalah n-heksan:etil asetat (5:1). Kristal putih yang diperoleh memiliki titik leleh 132-134oC. Berdasarkan data analisis UV, IR, NMR (1D dan 2D) dan diperkuat dengan HMQC, COSY dan HMBC, senyawa yang diperoleh diidentifikasi sebagai β-sitosterol. Pada pengujian aktivitas antidiabetes, senyawa murni β-sitosterol mempunyai aktivitas antidiabetes dengan IC50 723,9.
     
    The research that focused on the leaves of Durio zibethinus L. has been conducted. The aims of this research was to determine the chemical structure that has antidiabetic activity using α-glucosidase enzyme inhibition methods that presence in the leaves of Durio zibethinus L. This research was started by extracting durian leaves with 96% ethanol solvent. The concentrated ethanol extract was partitioned using nhexane, ethyl acetate and water. Furthermore, each fraction was subjected to phytochemical screening, and antidiabetic testing. The highest antidiabetic test results were obtained in the ethyl acetate fraction. Furthermore, the ethyl acetate fraction was isolated, purified, and then characterized using UV, IR, NMR (1D and 2D), and LCMS spectrophotometry methods. The ethyl acetate fraction was separated using column chromatography, the eluent that used in this process was n-hexane: ethyl acetate (5: 1). The obtained pure white crystal has a melting point of 132-134oC. Based on the several data, i.e. UV, IR, NMR (1D and 2D) and reinforced with HMQC, COSY and HMBC, the obtained compound was identified as β-sitosterol. In antidiabetic activity, the pure β-sitosterol compound has IC50 of 723,9.
