    Mikrostruktur Jaringan Tumor dan Ekspresi P53 pada Tikus Model Kanker Payudara yang Diberi Ekstrak Metanol Daun Biwa (Eriobotrya Japonica (Thunb.) Lindl.)

    Abstract
    Loquat (Eriobotrya japonica (Thunb.) Lindl) containing high antioxidant compound is suspected to have anticancer effect. This study aims to determine the effects of methanolic extract of loquat leaves on microstructre of tumor tissue and expression of p53 of breast cancer in rats induced by benzo(α)pyrene. The experiment used completely randomized design, in which rats divided into 5 group with 6 replicates each. The group are negatif control, without induced benzo(α)pyrene, positive control induced by benzo(α)pyrene at a dose of 50 mg/kg body weight (BW), and dose level administration were induced by benzo(α)pyrene at a dose of 50 mg/Kg body weight and given methanolic extract of loquat leaves consecutive dose 200; 300 and 400 mg/KgBW. Morphological observations showed that methanol extract of loquat leaves did not affect body weight and unable to decrease the diameter of tumor in rats. Histological observations showed that methanolic extract of loquat leaves were able to decrease the amount of vascularity in the mammary gland and there was an antiangiogenic effect by extract. The expression of p53 in dose level administration increased with the doses given, which indicate that the methanolic extract of loquat leaves has potential as an anticancer.
     
    Biwa (Eriobotrya japonica (Thunb.) Lindl) memiliki kandungan antioksidan tinggi yang diduga mempunyai efek penghambatan untuk karsinogenesis. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak metanol daun biwa terhadap mikrostruktur jaringan tumor dan ekspresi p53 pada tikus model kanker payudara yang diinduksi benzo(α)piren. Penelitian ini menggunakan rancangan acak lengkap, tikus dibagi menjadi 5 kelompok dengan 6 ulangan. Kelompok kontrol negatif, tanpa induksi benzo(α)piren, Kelompok kontrol positif, diinduksi benzo(α)piren dengan dosis 50mg/KgBB tikus tanpa pemberian ekstrak metanol daun biwa, kelompok pemberian dosis bertingkat diinduksi benzo(α)piren dengan dosis 50mg/KgBB tikus dan diberi dosis ekstrak metanol daun biwa berturut-turut 200; 300; dan 400 mg/KgBB tikus. Pengamatan morfologi menunjukkan bahwa ekstrak metanol daun biwa tidak dapat mempengaruhi peningkatan berat badan dan tidak mampu mengurangi diameter tumor. Pengamatan histologi menunjukkan bahwa ekstrak metanol daun biwa mampu menurunkan jumlah vaskularisasi pada kelenjar mammae dan terdapat efek antiangiogenik pada ekstrak metanol daun biwa. Ekspresi p53 pada kelompok pemberian dosis bertingkat meningkat seiring pemberian dosis yang diberikan, sehingga ekstrak metanol daun biwa berpotensi sebagai antikanker.
