dc.description.abstractOne of the causes of the increasing prevalence of Type 2 Diabetes Mellitus (T2DM) is lack of physical activity. Exercise is the main pillar in Diabetes management. The mechanism of glucose uptake mediated by exercise is different from insulin, and this mechanism is not disturbed in T2DM. One of the mechanisms is through the activation of 5'AMP-activated protein kinase (AMPK). AMPK not only increase the translocation of glucose transporter 4 (GLUT4), but also regulate GLUT4 expression. The most effective exercise strategy in T2DM has not been established. Aerobic exercise consists of continuous and interval training. This study aims to determine the effect of various physical exercise models on the expression of Ampk α2 and Glut 4 in skeletal muscle. This research is a true experimental with a posttest design with a control group. This study uses skeletal muscles stored biological material of 20 rat Wistar T2DM divided into K1 (control) not given exercise, K2 moderate continuous training, K3 severe continuous training, K4 slow interval training, and K5. fast interval training. Running on a treadmill frequency 3 times a week for 8 weeks. The relative expression of Ampk α2 and Glut 4 was assessed by the Real Time PCR method and the results were compared between the treatment group and the control group. Data analysis using Kruskal Wallis and Livak formula. The values of ΔCt Ampk α2 and Glut 4 in each group were not significantly different (p = 0.309 and p = 0.909). From the Livak formula, it was found that the largest increase in expression occurred in K5 with an increase in the expression of Ampk α2 4.56 times and the expression of Glut 4 increased 4.79 times compared to K1. Conclusion: continuous and interval training increase the expression of Ampk α2 and Glut 4. The greatest increase in expression occurs in fast interval training.en_US
dc.description.abstractSalah satu penyebab peningkatan prevalensi Diabetes Mellitus Tipe 2 (DM Tipe-2) adalah penurunan aktivitas fisik. Olahraga menjadi pilar utama dalam pengelolaan Diabetes. Mekanisme pengambilan glukosa yang dimediasi oleh olahraga berbeda dengan insulin, dan mekanisme tersebut tidak terganggu pada penderita DM Tipe- 2. Salah satu mekanismenya adalah melalui aktivasi 5’AMP-activated protein kinase (AMPK). AMPK tidak hanya dapat meningkatkan translokasi glucose transporter 4 (GLUT4), tetapi juga meregulasi ekspresi GLUT4. Strategi olahraga yang paling efektif pada DM Tipe-2 belum ditetapkan. Olahraga aerobik terdiri dari tipe continuous dan interval. Penelitian ini bertujuan untuk mengetahui pengaruh berbagai model latihan fisik terhadap ekspresi Ampk α2 dan Glut 4 di otot rangka. Penelitian ini merupakan penelitian eksperimental murni dengan desain penelitian posttest with control group. Penelitian ini menggunakan bahan biologi tersimpan berupa otot rangka tikus wistar model DM Tipe-2 sebanyak 25 ekor yang dibagi atas K1 (kontrol) tidak diberikan latihan, K2 latihan continuous intensitas sedang, K3 latihan continuous intensitas berat, K4 latihan interval tipe lambat, dan K5 latihan interval tipe cepat. Latihan berupa berlari di atas treadmill frekuensi 3 kali seminggu selama 8 minggu. Ekspresi Ampk α2 dan Glut 4 dinilai dengan metode Real Time PCR. Analisis data menggunakan uji one way Anova, Kruskal Wallis, dan rumus Livak. Nilai ΔCt Ampk α2 dan Glut 4 pada tiap kelompok tidak berbeda bermakna (p= 0,737 dan p= 0,909). Dari rumus Livak didapat peningkatan ekspresi terbesar terjadi pada K5 dengan peningkatan ekspresi Ampk α2 4,56 kali dan ekspresi Glut 4 meningkat 4,79 kali dibandingkan K1. Kesimpulan: latihan tipe continuous dan interval meningkatkan ekspresi Ampk α2 dan Glut 4. Peningkatan ekspresi terbesar terjadi pada latihan interval tipe cepat.en_US
