Aktivitas Antioksidan Ekstrak Daun Pirdot (Saurauia vulcani Korth.) dan Efeknya terhadap Mikrostruktur Ginjal Mencit (Mus musculus L.) Model Diabetes Tipe 2

Abstract

Pirdot merupakan salah satu tumbuhan endemik Sumatera Utara. Beberapa penelitian menunjukkan bahwa ekstrak daun pirdot mampu menurunkan kadar gula darah pada penderita diabetes tipe 2. Tujuan dari penelitian ini adalah untuk mengetahui aktivitas antioksidan (IC50) ekstrak daun pirdot, efek ekstrak terhadap berat badan, dan efek terhadap morfometri dan mikrostruktur ginjal mencit model diabetes tipe 2. Metode penelitian menggunakan rancangan acak lengkap (RAL) yang terdiri atas lima perlakuan dengan masing-masing 5 ulangan. Penelitian ini terdiri dari kelompok kontrol negatif (K-) yaitu mencit normal, kontrol positif (K+) disuntik streptozotocin 50 mg/kg BB, dan tiga kelompok yang disuntik streptozotocin (P1, P2, P3) yang berturut-turut diberi ekstrak daun pirdot 100, 200, atau 300 mg/kg BB. Mencit dibedah kemudian diambil organ ginjalnya. Kemudian dilakukan pembuatan preparat organ ginjal dengan metode paraffin dan pewarnaan Hematoksilin-Eosin (HE). Hasil uji antioksidan menunjukkan bahwa ekstrak etanol daun pirdot memiliki nilai IC50 sebesar 22,37 μg/ml yang dapat dinyatakan sebagai antioksidan yang sangat kuat. Pemberian ekstrak etanol daun pirdot dosis 200 mg/kg BB mampu menaikan berat badan mencit yang telah kehilangan berat badan setelah disuntikkan STZ (p<0,05) dan menunjukkan adanya pengaruh (p<0,05) terhadap ukuran ginjal, penurunan ketebalan membran basalis dan jumlah sel mesangial mencit diabetes tipe 2.

The tratments consisted of consisted of a negative control group (K-), namely normal mice, positive control (K +) injected with streptozotocin 50 mg / kg BW, and three groups that were injected with streptozotocin (P1, P2, P3) which were consecutively given 100 pirdot leaf extract. , 200, or 300 mg / kg BW. The mice were dissected and the kidneys were taken and processed for routine histological observation. The result shows, the antioxidant activity of the ethanol extract of pirdot leaves had an IC50 value of 22.37 μg / ml which could be stated as a very strong antioxidant. The administration of pirdot leaf ethanol extract at a dose of 200 mg/kg BW was able to increase the bodyweight of mice that had lost weight after being injected with STZ (p <0.05) and showed an effect (p <0.05) on kidney size, decreased basement membrane thickness and the number of mesangial cells in type 2 diabetic mice.
