    Pengaruh Pemberian Vitamin E terhadap Gambaran Histologis Testis dan Jumlah Sel Sperma Mencit (Mus musculus, L.) yang Dipapari Tuak

    Abstract
    Vitamin E is one of the fat-soluble vitamins. Vitamin E acts as an antioxidant and can protect the biological action of membrane damage by free radicals. A free radicals is an atom or a molecule without a pair of electrons that can damage the molecules essential for cellular function. Giving antioxidants in the form of vitamin E is proposed to reduce the effects of free radicals in the body. This study aims to determine the effect of vitamin E on testicular histological features, the number of Leydig cells and testicular sperm count in mice given palm wine. In this study 30 healty and fertile adult male mice (Mus musculus, L.) strain DD Webster. The 8- 11 week old mice (20-35 g) were divided in to 6 treatment groups. The first group (K0) = consisted of 5 mice without treatment for 30 days. The first treatment group (P1) consisted of 5 mice that were each given palm wine (20% alcohol) orally at 0.5 ml/day for 15 days. After 15 the palm wine was repleaced with water 0,5 ml. The second treatment group (P2) consisted of 5 mice that were each given palm wine (20% alcohol) orally 0.5 ml/day for 30 days. The third treatment group (P3) consisted of 5 mice that were each given palm wine (20% alcohol) orally 0.5 ml/day for 15 days. After 15 days the palm wine was replaced with 0.25 mg of vitamin E/day administered orally. The fourth treatment group (P4) consisted of 5 mice that were each given palm wine (20% alcohol) orally 0.5 ml/day for 30 days. After 15 days in addition to the palm wine with 0.25 mg of vitamin E/day was administered orally. The fifth treatment group (P5) consisted of 5 mice that were each given palm wine (20% alcohol) 0.5 ml/day and vitamin E 0.25 mg/day orally. The mice were placed into groups randomly. This study was approved by the USU research ethics committee. The results of this study suggest concurrent administration of vitamin E (0.25 mg/day) reduces the negative affect of palm wine. An increased number of Leydig cells, an increased diameter of seminiferous tubules and an increased number spermatozoa were observed in treatment group P5 as compared to the other treatment groups.
     
    Vitamin E berperan sebagai antioksidan dan dapat melindungi aksi kerusakan membran biologis akibat radikal bebas. Radikal bebas adalah suatu atom dan molekul yang tidak mempunyai pasangan elektron dan dapat merusak molekul-molekul penting bagi fungsi seluler. Pemberian asupan antioksidan berupa vitamin E diusulkan dapat menurunkan efek radikal bebas dalam tubuh. Penelitian ini bertujuan untuk mengetahui tentang pengaruh pemberian vitamin E terhadap gambaran histologis testis, jumlah sel Leydig dan jumlah sperma pada testis mencit yang di papari tuak. Pada penelitian ini menggunakan mencit jantan (Mus musculus, L) strain DD Webster dewasa sehat dan fertil yang berumur 8-11 minggu dengan berat 20-35 g sebanyak 30 ekor dibagi dengan 6 kelompok perlakuan. Kelompok (K0) = kelompok kontrol pertama terdiri dari 5 ekor mencit dewasa jantan tanpa perlakuan selama 30 hari. Kelompok 2 (P1) = Kelompok perlakuan pertama terdiri dari 5 ekor mencit dewasa jantan yang diberi tuak (alkohol 20%) 0,5 ml/hari/ekor secara oral setiap hari selama 15 hari pertama dan 15 hari berikutnya pemberian tuak dihentikan dan diganti dengan pemberian aquadest 0,5 ml. Kelompok 3 (P2) = Kelompok perlakuan kedua terdiri 5 ekor mencit dewasa yang diberi tuak (alkohol 20%) 0,5 ml/hari/ekor secara oral selama 30 hari. Kelompok 4 (P3) = Kelompok perlakuan ketiga terdiri 5 ekor mencit dewasa yang diberi tuak (alkohol 20%) 0,5 ml /hari/ekor selama 15 hari pertama dan 15 hari berikutnya pemberian tuak dihentikan diganti dengan pemberian vitamin E 0,25 mg/hari/ekor/mencit secara oral. Kelompok 5 (P4) = Kelompok perlakuan keempat terdiri 5 ekor mencit dewasa yang diberi tuak (alkohol 20%) 0,5 ml/hari/ekor selama 15 hari pertama dan 15 hari berikutnya pemberian tuak dengan pemberian vitamin E 0,25 mg/ekor/hari secara oral. Kelompok 6 (P5) = Kelompok perlakuan kelima terdiri dari 5 ekor mencit dewasa yang diberi tuak (alkohol 20%) 0,5 ml/hari/ekor dan pemberian vitamin E 0,25 mg/ekor/hari selama 30 hari secara oral. Mencit ditempatkan ke dalam kelompok secara random. Penelitian ini telah mendapat persetujuan dari komite etik penelitian USU. Hasil yang didapat pemberian vitamin E 0,25 mg/hari/mencit sejalan dengan pemaparan tuak selama 30 hari cenderung mempengaruhi peningkatan jumlah sel Leydig mencit, peningkatan diameter tubulus seminiferus mencit dan peningkatan jumlah spermatozoa mencit.
