    Pengaruh Pemberian Curcuminoid dalam Mencegah dan Memperbaiki Kerusakan Fibroblas Koklea Rattus Norvegicus Model Diabetes Mellitus ditinjau dari Ekspresi Kolagen Tipe IV

    Abstract
    Introduction: Damage to fibroblasts within the cochlear lateral wall is suggested as one of the causes of hearing loss in patients with diabetes mellitus. Aim: The aim of this study was to prove curcuminoid as a safe and effective substance to prevention and treat the fibroblasts within cochlear lateral wall damage measured by the expressions of type IV collagen. Method: Twenty-four Rattus norvegicus were randomly divided into 6 groups (n=4). Group 1: the control group; group 2: the diabetic group; group 3 and 4: the diabetic groups received curcuminoid therapy 200 and 400 mg/kgBW/day from day 3 - 5; group 5 and 6: the diabetic groups received curcuminoid therapy 200 and 400 mg/kgBW/day from day 3 - 10. Curcuminoid [16.62 ± 0.14]% w/w compared to Standard was administered orally, derived from Curcuma longa Linnaeus (turmeric). All samples were immunohistochemistrically examined for the expressions of type IV collagen in fibroblasts within cochlear lateral wall. Result: Statistically significant differences were found for the expressions of type IV collagen (p<0.05) between group 1 and 2, group 2 and 5, and group 2 and 6. Conclusion: Curcuminoid proved to be potentially effective in the treatment of damage to fibroblasts within cochlear lateral wall regarding the increased expression of type IV collagen in diabetic rat models
     
    Pendahuluan: Kerusakan pada dinding lateral koklea dianggap salah satu faktor penyebab gangguan pendengaran akibat diabetes mellitus. Tujuan Penelitian: Penelitian ini bertujuan untuk membuktikan bahwa curcuminoid adalah zat yang aman dan efektif untuk mencegah dan memperbaiki kerusakan fibroblas dinding lateral koklea ditinjau dari ekspresi kolagen tipe IV. Metode: Penelitian ini terdiri dari 24 tikusWistar Rattus norvegicus yang dipilih secara acak menjadi 6 kelompok (n=4). Kelompok 1: kontrol; kelompok 2: diabetes; kelompok 3 dan 4: diabetes + curcuminoid 200 dan 400 mg/kgbb/hari dari hari ketiga sampai hari kelima; kelompok 5 dan 6: diabetes + curcuminoid 200 dan 400 mg/kgbb/hari dari hari ketiga sampai hari kesepuluh. Curcuminoid [16.62 ± 0.14]% b/b yang diberikan secara oral, berasal dari Curcuma longa Linnaeus (kunyit). Semua sampel dilakukan pemeriksaan immunohistokimia untuk menilai ekspresi kolagen tipe IV pada fibroblas dinding lateral koklea. Hasil Penelitian: Terdapat perbedaan ekspresi kolagen tipe IV yang signifikan secara statistik (p<0,05) antara kelompok 1 dengan 2, kelompok 2 dengan 5 dan kelompok 2 dengan 6. Kesimpulan: Curcuminoid terbukti berpotensi efektif dalam mencegah dan memperbaiki kerusakan fibroblas dinding lateral koklea dengan meningkatkan ekspresi kolagen tipe IV pada tikus model diabetes.
