    Pengendalian Biofilm Streptococcus Agalactiae pada Permukaan Sisik Ikan dan Plastik Pvc dengan Senyawa Antibakteri Bakteri Asam Laktat Perairan Tawar

    Abstract
    Streptococcus agalactiae is bacteria that cause streptococcosis on fish and able to form biofilm on solid surface. The aim of this research is to control biofilm S.agalactiae by using antibacteria compound of Lactid Acid Bacteria (LAB) isolated from fresh water of Nila fish pond around Medan city, North Sumatera. There were 21 LAB isolated and 4 of them were able to inhibit S. agalactia. The selected isolate was AK 5 as Lactobacillus plantarum that found as potential biocontrol agent which showed the higher inhibition compare to other 21 isolate (17,5mm). L. plantarum was used to produce antibacterial compound to control biofilm cells of S. agalactiae on nila fish scale and polyvinyl chloride (PVC). Biofilm formation of S. agalactiae was perform in 1, 3 and 5 days in Nutrion Broth media on fish scale and PVC then were detached with micro glass bead for counting. The highest number of biofilm cells were found at 3 days incubation as much as 106 CFU/chip both on plastic and fish scale surfaces when perform together but they were 105 and 104 CFU/chip of biofilm cells found in each surface. Controlling of 3 days-age biofilm cell of S. agalactiae with 100% crude extract of antibacterial compound of LAB isolate reduced 101 CFU/chip biofilm cell of S. agalactiae. Antibactery compounds was tasted to saw its resitance to pH and surfactant. Antibactery compounds of LAB selected was relatively stable at pH 2 to pH 10 and at temperature 40 0C,60 0C,80 0 Keyword : Lactid Acid Bacteria (LAB), L. plantarum, S. agalactiae, biofilm, scales and plastic. C for S. agalactiae. Addition EDTA into crude extract of antibacterial compound L. plantarum showed that not any inhibition zone of S. agalactiae, whereas additional tween 80 made reduction inhibition zone. conclude of this research is antibateria compound crude extract L. plantarum able to inhibit biofilm cells.
     
    Streptococcus agalactiae merupakan bakteri penyebab streptococosis pada ikan dan mampu membentuk biofilm pada permukaan padat. Penelitian ini bertujuan untuk mengendalikan biofilm S agalactiae dengan memanfaatkan senyawa antibakteri bakteri asam laktat (BAL) yang diisolasi dari air kolam ikan nila Sumatera Utara. Dari 21 isolat hasil isolasi pada media MRSA didapatkan isolat AK 5 yang teridentifikasi sebagai Lactobacillus plantarum paling potensial dalam menghambat S.agalactiae dengan diameter zona hambat 17,5 mm. Perhitungan jumlah sel dan hasil pengendalian biofilm pada sisik ikan nila dan plastik PVC dilakukan dengan metode pelepasan sel biofilm dengan serbuk kaca halus (glass bead), kemudian kultur disebar pada cawan untuk perhitungan cawan total (TPC). Pembentukan biofilm dilakukan pada hari ke 1, 3 dan 5. Biofilm S.agalactiae pada lempeng yang berada pada tabung terpisah mampu membentuk biofilm dengan jumlah sel tertinggi pada hari ke-3 yaitu sebesar 106 CFU/lempeng baik pada permukaan lempeng sisik ikan maupun plastik PVC. Biofilm S.agalactiae pada lempeng yang berada dalam satu tabung yang sama mampu membentuk biofilm lebih besar pada permukaan lempeng plastik yaitu 105 CFU/lempeng sedangkan pada permukaan lempeng sisik sebesar 104 CFU/lempeng. Senyawa antibakteri ekstrak kasar L. plantarum mampu menurunkan jumlah sel biofilm pada masing-masing uji sebesar 101 CFU/lempeng. Senyawa antibakteri L. plantarum stabil pada pH 2-10 dan suhu 40, 60 dan 80 0 C. EDTA mampu merusak aktivitas senyawa antibakteri L. plantarum dan tween 80 tidak mampu menurunkan aktivitas senyawa tersebut. Dari hasil penelitian dapat disimpulkan bahwa senyawa antibakteri ekstrak kasar ekstrak kasar L. plantarum dapat menghambat sel biofilm S. agalactiae.
