    Isolasi Bakteri Penghasil Antimikroba dari Sampel Tanah Taman Wisata Alam Sibolangit dan Kemampuannya Menghambat Pertumbuhan Mikroba Patogen

    Abstract
    A study on isolation and assay of antimicrobial activity of soil bacterial isolates of TWA Sibolangit, North Sumatra has been done. Nutrient agar was used in microbial isolation. Twenty-nine isolates showed different potential in inhibiting pathogens growth. Methanol, n-hexane, and ethyl acetate extraction method was utilised to get antimicrobial metabolites. Antagonistic test of the three selected potential extract was done using disc diffusion methods against Escherichia coli, Staphylococcus aureus, Candida albicans, Fusarium oxysporum, and Ganoderma boninense. In vitro test of the extract showed that methanol and n-hexane extract of S3T32-3 were active against E. coli, C. albicans, and G. boninense, but methanol to inhibit S. aureus. Methanol and n-hexane extract of S3T32-3 showed the same activity against F. oxysporum. Ethyl acetate S3T33-3 extract showed the lowest activity against G. boninense.
     
    Telah dilakukan penelitian tentang isolasi bakteri tanah yang berpotensi menghasilkan antimikroba dari Taman Wisata Alam Sibolangit, Sumatera Utara. Media nutrient agar digunakan untuk isolasi awal. Dua puluh sembilan isolat yang diperoleh memiliki kemampuan yang berbeda terhadap mikroba uji. Metanol, nheksana, dan etil asetat digunakan sebagai pelarut untuk memperoleh ekstrak metabolit. Pengujian antagonis ekstrak tiga isolat potensial terpilih dilakukan dengan metode difusi cakram terhadap mikroba uji Escherichia coli, Staphylococcus aureus, Candida albicans, Fusarium oxysporum, dan Ganoderma boninense. Hasil pengujian secara in vitro menunjukkan bahwa ekstrak metanol dan n-heksana isolat S3T33-3 aktif terhadap E. coli, C. albicans, dan G. boninense, sedangkan terhadap S. aureus hanya metanol. Ekstrak menggunakan pelarut metanol dan n-heksana isolat S3T32-3 menunjukkan aktivitas yang sama terhadap F. oxysporum. Ekstrak etil asetat isolat S3T33-3 menunjukkan aktifitas yang rendah terhadap G. boninense.
