dc.description.abstract | Mangosteen peel extract is now widely used as an alternative treatment by society as an anti-oxidant without knowing the side effects. The consumption of Monosodium Glutamate (MSG) as food additive can cause various side effects due to the formation of free radicals in body such as liver dysfunction. Vitamin E is widely used as anti-oxidants to prevent free radical communities. The research method uses true experimental design and aims to compare the effect of ethanol extract of mangosteen peel (EEKM) on the damage of liver that is caused by MSG giving to vitamin E. From 20 samples of mice, DDW strain male mice divides into four treatment groups, namely; control group / distilled water (P0), MSG giving group (P2), the giving of MSG and EEKM(P3) group and the last is MSG group and vitamin E (P4) . Thus, some assessments are done to the liver function (serum levels of AST / ALT) and macroscopic together with microscopic changing in the liver of mice with heamatoksilin eosin staining. As well as evaluating the damage level of immunohistochemical staining with TNF-α (NBP1-67 821, pAb, 1:50 dilution). The giving of Ethanol extract of Mangosteen peel has the same power with vitamin E (p> 0.05) in lowering AST, except in serum ALT levels (p <0,05). The Macroscopic and microscopic assessment of mice after giving EEKM and vitamin E did not show any significant differences in fixing the damage of liver from MSG giving (p> 0,05). Ethanol extract of mangosteen peel and vitamin E both are able to improve the look of immunohistochemical expression of TNF-α inflammatory process that is caused by MSG giving (p>0,05). | en_US |
dc.description.abstract | Ekstrak kulit manggis saat ini banyak digunakan sebagai alternatif pengobatan oleh masyarakat sebagai anti-oksidan tanpa mengetahui efek sampingnya. Konsumsi Monosodium glutamate (MSG) yang digunakan sebagai penambah rasa makanan di masyarakat dapat menimbulkan berbagai efek samping karena terbentuknya radikal bebas di tubuh antara lain gangguan fungsi hati. Vitamin E yang selama ini dikenal sebagai anti-oksidan banyak digunakan masyarakat untuk mencegah radikal bebas. Metode penelitian menggunakan true experimental designs yang bertujuan membandingkan pengaruh pemberian ekstrak etanol kulit manggis (EEKM) pada kerusakan hati akibat pemberian MSG terhadap pemberian vitamin E. Dari 20 ekor sampel mencit jantan strain DDW, yang dibagi atas 4 kelompok perlakuan, yaitu kelompok kontrol/aquadest (P0), kelompok pemberian MSG (P2), kelompok pemberian MSG dan EEKM (P3), dan kelompok MSG dan vitamin E (P4). Kemudian dinilai fungsi hati (kadar serum AST/ALT), perubahan makroskopik dan mikroskopik hati mencit dengan pewarnaan heamatoksilin eosin, serta penilaian tingkat kerusakan hati dengan pewarnaan imunohistokimia TNF-α(NBP1-67821, pAb, pengenceran 1:50). Pemberian ekstrak etanol kulit manggis mempunyai kemampuan yang sama dengan vitamin E (p>0,05) dalam menurunkan AST, kecuali dalam menurunkan kadar serum ALT(p<0,05). Penilaian makroskopik dan mikroskopik hati mencit setelah pemberian EEKM dan vitamin E tidak menunjukkan perbedaan yang bermakna dalam memperbaiki kerusakan hati akibat pemberian MSG (p>0,05). Ekstrak etanol kulit manggis dan vitamin E sama-sama dapat memperbaiki tampilan imunohistokimia ekspresi proses inflamasi TNF-α akibat pemberian MSG (p>0,05). | en_US |
