dc.description.abstractObjective: The aim of this study was to investigate the potential of curcumin as an antioxidant to increase the expressions of SOD in fibroblasts of the cochlear lateral wall in rat models of diabetes mellitus. Materials and Methods: Twenty-four male Wistar rats Rattus norvegicus were randomly divided into six groups: Group 1 as the control group; Group 2 as the diabetic group; Group 3 and 4 as the diabetic groups received curcumin therapy 200 and 400 mg/kg b.w. for 3 days, respectively; Group 5 and 6 as the diabetic groups received curcumin therapy 200 and 400 mg/kg b.w. for 8 days, respectively. Induction of diabetes using injection of streptozotocin 60 mg / kg b.w intraperitoneally. The curcuminoid used was obtained from Curcuma longa L. with curcuminoid content of 28.1 ± 1.0)% w / w compared to the standard. All rats underwent termination and necropsy procedure on their temporal bones for immunohistochemical assay to determine the expressions of SOD. Results : The decreased expression of SOD was detected in the diabetic group (without curcumin treatment). The treatment of curcumin at doses of 200 and 400 mg/kg b.w. for 3 and 8 days led to significant differences (p < 0.05) in the expressions of the SOD compared to diabetic group (without curcumin treatment). No significant differences were found in terms of dose and duration of curcumin administration on the expressions of SOD. Conclusion : Curcumin may act as an antioxidant against oxidative stress due to diabetes mellitus via increased expressions of SOD on cochlear fibroblasts in rat models of diabetes mellitus.en_US
dc.description.abstractTujuan: Penelitian ini bertujuan untuk melihat potensi curcuminoid sebagai antioksidan untuk meningkatkan ekspresi super oxide dismutase (SOD) pada fibroblas dinding lateral koklea pada tikus model diabetes mellitus. Metode: Dua puluh empat tikus jantan Rattus norvegicus galur wistar dibagi secara acak menjadi enam kelompok: Kelompok 1 : kelompok kontrol, kelompok 2 : kelompok model diabetes, kelompok 3 dan 4 kelompok diabetes yang mendapatkan terapi curcuminoid masing – masing 200 dan 400 mg / kgbb selama 5 hari, kelompok 5 dan 6 kelompok diabetes yang mendapatkan terapi curcuminoid masing – masing 200 dan 400 mg/kg bb selama 10 hari. Induksi diabetes menggunakan injeksi streptozotocin 60 mg/kg bb intraperitoneal. Curcuminoid yang digunakan berasal dari Curcuma longa L. dengan kadar curcuminoid 28,1 ± 1,0)% b/b dibandingkan standar. Semua tikus diterminasi dan dilakukan nekropsi pada tulang temporal seluruh sampel dan dilakukan pemeriksaan imunohistokimia untuk menentukan ekspresi SOD. Hasil: Terjadi penurunan ekspresi SOD pada kelompok model diabetes tanpa pemberian curcuminoid. Pemberian curcuminoid dengan dosis 200 dan 400 mg/kg bb selama 5 dan 10 hari menyebabkan perbedaan ekspresi SOD yang bermakna secara statistik (p < 0.05) dibandingkan dengan kelompok model diabetes tanpa pemberian curcuminoid. Tidak dijumpai perbedaan yang bermakna dalam hal perbedaan dosis dan durasi pemberian curcuminoid terhadap ekspresi SOD. Kesimpulan: Curcuminoid dapat bekerja sebagai antioksidan dalam keadaan stres oksidatif akibat diabetes melitus melalui peningkatan ekspresi SOD pada fibroblas koklea pada model tikus diabetes mellitus.en_US
