    Pengaruh Pemberian Curcuminoid terhadap Konsentrasi Hidrogen Peroksida (H2O2) Serum dan Ekspresi Malondialdehid (MDA) Fibroblas Koklea pada Rattus Norvegicus Model Diabetes Mellitus

    Abstract
    Objective: To investigate the potential of curcuminoid as an antioxidant to reduce concentration of serum hydrogen peroxide (H2O2) and the expressions of malondialdehyde (MDA) in fibroblasts of the cochlear lateral wall in rat models of diabetes mellitus. Methods: Twenty-four male Wistar rats Rattus norvegicus were randomly divided into six groups: Group 1 as the control group; Group 2 as the diabetic group; Group 3 and 4 as the diabetic groups received curcumin therapy 200 and 400 mg/kg b.w. for 5 days, respectively; Group 5 and 6 as the diabetic groups received curcumin therapy 200 and 400 mg/kg b.w. for 10 days, respectively. Induction of diabetes using injection of streptozotocin 60 mg / kg b.w intraperitoneally. The curcuminoid used was obtained from Curcuma longa L. with curcuminoid content of 28.1 ± 1.0)% w / w compared to the standard. Precordial blood samples, and temporal bones tissues were taken for ELISA and immunohistochemical assay, respectively, to determine concentration of serum H2O2 and the expressions of malondyaldehyde (MDA) in fibroblasts of the cochlear lateral wall, respectively. Results: The increased of concentration of serum H2O2 and the expressions of MDA in fibroblasts of the cochlear lateral wall were detected in the diabetic group (without curcuminoid treatment). The treatment of curcuminoid at doses of 400 mg/kg b.w. for 3 and 8 days led to significant differences (p < 0.05) in the concentration of serum H2O2 and the expressions of MDA in fibroblasts of the cochlear lateral wall compared to diabetic group (without curcuminoid treatment). Conclusion: Curcuminoid has the ability to attenuate free radicals produced by the state of hyperglycemia, either in the form of H2O2 in serum, or MDA in cochlear tissues of lateral wall in rat model of diabetes mellitus.
     
    Tujuan: Penelitian ini bertujuan untuk melihat potensi curcuminoid sebagai antioksidan untuk menurunkan konsentrasi hidrogen peroksida (H2O2) serum dan ekspresi malondialdehid (MDA) fibroblas koklea pada tikus model diabetes mellitus. Metode: Dua puluh empat tikus jantan Rattus norvegicus galur wistar dibagi secara acak menjadi enam kelompok: Kelompok 1 : kelompok kontrol, kelompok 2 : kelompok model diabetes, kelompok 3 dan 4 kelompok diabetes yang mendapatkan terapi curcuminoid masing – masing 200 dan 400 mg / kgbb selama 5 hari, kelompok 5 dan 6 kelompok diabetes yang mendapatkan terapi curcuminoid masing – masing 200 dan 400 mg/kg bb selama 10 hari. Induksi diabetes menggunakan injeksi streptozotocin 60 mg/kg bb intraperitoneal. Curcuminoid yang digunakan berasal dari Curcuma longa L. dengan kadar curcuminoid 28,1 ± 1,0)% b/b dibandingkan standar. Semua tikus diterminasi dan diambil darah prekordial serta dilakukan nekropsi pada tulang temporal seluruh sampel dan dilakukan pemeriksaan ELISA untuk menentukan konsentrasi H2O2 serum dan imunohistokimia untuk melihat ekspresi MDA. Hasil: Terjadi peningkatan konsentrasi H2O2 serum dan ekspresi MDA fibroblas koklea pada kelompok model diabetes tanpa pemberian curcuminoid. Pemberian curcuminoid dengan dosis 400 mg/kg bb selama 5 dan 10 hari menyebabkan perbedaan konsentrasi H2O2 serum dan ekspresi MDA fibroblas koklea yang bermakna secara statistik (p < 0.05) dibandingkan dengan kelompok model diabetes tanpa pemberian curcuminoid. Kesimpulan: Curcuminoid memiliki kemampuan untuk menurunkan radikal bebas pada keadaan hiperglikemia, baik berupa H2O2 serum, atau MDA dalam fibroblas dinding lateral koklea pada tikus model diabetes mellitus.
