Efek Ekstrak Daun Pirdot (Saurauia Vulcani K.) terhadap Ekspresi Insulin Pankreas Mencit (Mus musculus L.) Model Diabetes Tipe 2

Abstract

Research on the effect of pirdot leaf extract (Saurauia vulcani K.) on the insulin expression of mice (Mus musculus L.) pancreas type 2 diabetes model has been carried out from June 2020 to November 2020. The pirdot plant (Saurauia vulcani K.) was used by the community for the treatment of diabetes. Pirdot leaves have flavonoid compounds, which were able to lower blood sugar levels and could reduce the level of damage to the pancreas. The study aimed to determine the effect of ethanol extract of pirdot leaves on blood sugar levels using the glucose tolerance test, insulin expression, and pancreatic damage of type 2 diabetic mice. This study used a completely randomized design (CRD) consisting of 5 treatments, negative control, positive control, treatment with ethanol extract of pirdot leaves 100 mg/Kg BB, 200 mg/Kg BB and 300 mg/Kg BB. The treatment was carried out for 35 days and then surgery was carried out and the pancreas was taken to make histological preparations using the paraffin method with Hematoxylin-Eosin (HE) staining to see the level of qualitative damage to the pancreas and Immunohistochemical (IHC) staining to see the insulin expression of β cells of the Langerhans islets. The results showed that the ethanol extract of pirdot leaves with doses of 100 mg/kg BB is the optimum doses had the effect of reducing blood sugar levels in mice that were induced by glucose orally, could increase the insulin expression of β cells of the Langerhans islets and could reduce the level of β cell damage of the pancreatic Langerhans islets.

Penelitian mengenai efek ekstrak daun pirdot (Saurauia vulcani K.) terhadap ekspresi insulin pankreas mencit (Mus musculus L.) model diabetes tipe 2 telah dilakukan pada bulan Juni 2020 sampai dengan November 2020. Tanaman pirdot (Saurauia vulcani K.) dimanfaatkan masyarakat untuk pengobatan diabetes. Daun pirdot memiliki senyawa flavonoid, mampu menurunkan kadar gula darah dan dapat mengurangi tingkat kerusakan pada pankreas. Penelitian bertujuan untuk mengetahui pengaruh ekstrak etanol daun pirdot terhadap pola penurunan kadar gula darah setelah diberikan glukosa secara oral, ekspresi insulin dan kerusakan pankreas pada mencit diabetes tipe 2. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) yang terdiri dari 5 perlakuan, kontrol negatif, kontrol positif, perlakuan ekstrak etanol daun pirdot dosis 100 mg/Kg BB, 200 mg/Kg BB, dan 300 mg/Kg BB. Perlakuan dilakukan selama 35 hari dan kemudian dilakukan pembedahan dan diambil organ pankreas untuk dibuat preparat histologi dengan metode paraffin dengan pewarnaan Hematoksilin-Eosin (HE) untuk melihat tingkat kerusakan kualitatif pankreas dan pewarnaan Immunohistokimia (IHC) untuk melihat ekspresi insulin sel β pulau Langerhans. Hasil penelitian menunjukkan bahwa ekstrak etanol daun pirdot dengan dosis 100 merupakan dosis optimum memiliki efek penurunan kadar gula darah pada mencit yang diinduksi glukosa secara oral, meningkatkan ekspresi insulin sel β pulau Langerhans dan mengurangi tingkat kerusakan sel β pulau Langerhans pankreas.
