dc.description.abstractKontaminasi dan serangan cendawan termasuk Aspergillus flavus hampir menyerang seluruh komoditas pertanian dan perkebunan dan salah satu di antaranya adalah biji kopi. Serangan cendawan tidak hanya merusak secara fisik namun juga menghasilkan mikotoksin khususnya aflatoksin yang berbahaya bagi manusia dan hewan apabila terkonsumsi. Kontaminasi cendawan dapat terjadi saat prapanen, ketika biji masih di kebun maupun saat pascapanen seperti cara penjemuran ataupun penyimpanan yang kurang tepat. Penelitian ini bertujuan menganalisis serangan cendawan dan toksigenitas galur-galur A. flavus pada lahan perkebunan dan biji kopi pada perkebunan kopi rakyat di Berastagi, Kabupaten Karo, Sumatera Utara. Populasi cendawan ditentukan berdasarkan pengenceran berderet dilanjutkan dengan cawan tuang pada medium Dichloran 18% Glycerol Agar (DG18). Uji toksigenitas galur-galur A. flavus secara kualitatif dilakukan dengan mengkultur pada medium Coconut Agar Medium (CAM) dan secara semi-kuantitatif menggunakan kromatografi lapis tipis. Hasil penelitian ini menunjukkan bahwa terdapat 11 spesies cendawan tanah yang berhasil diisolasi, terutama didominasi oleh genus Aspergillus. Biji kopi yang sedang dijemur terserang oleh 4 spesies cendawan yaitu, A. terreus, A. repens, Cladosporium sp. dan Mucor sp. Biji kopi kering simpan terserang oleh 4 spesies yaitu A. chevalieri, A. niger, Penicillium citrinum, Rhizopus sp. Sebanyak 11 galur A. flavus berhasil diisolasi di antaranya 3 galur A. flavus dari tanah perkebunan kopi, 7 galur A. flavus dari biji kopi yang sedang dijemur dan 1 galur A. flavus yang dari biji kopi kering simpan. Berdasarkan uji toksigenitas ditemukan bahwa 10 galur A. flavus bersifat toksigen yaitu 3 galur dari tanah perkebunan kopi, 6 galur A. flavus dari biji yang sedang disimpan, 1 galur dari biji kering simpan. Sebanyak 1 galur A. flavus bersifat non-toksigen yang diisolasi dari biji kopi yang sedang dijemur. Analisis kadar aflatoksin menunjukkan galur A. flavus yang diisolasi dari tanah perkebunan kopi menghasilkan kadar aflatoksin tertinggi yaitu 42,4 μg/kg.en_US
dc.description.abstractFungal contamination and Aspergillus flavus is commonly contaminate to all crop commodities including coffee beans. Fungal contamination on coffee beans not only physically damaging but some fungal species produce mycotoxins that harmful to humans and animals. Fungal contamination on coffee beans occured during preharvest, when the commodity in the field or during post-harvest handling such as drying or storage. This study aimed to analyze fungal contamination and toxigenicity of A. flavus strains on coffee plantations and coffee beans in smallholder coffee plantations in Berastagi, Karo Regency, North Sumatera. The fungal population was determined based on serial dilution followed by pour plate on Dichloran 18% Glycerol Agar (DG18) medium. Toxigenicity of A. flavus strains was conducted qualitatively by culturing on Coconut Agar Medium (CAM) and semi-quantitatively aflatoxin was carried out using thin layer chromatography. The results showed that a total of 11 species of soil fungi on the plantation area were isolated, mainly dominated by Aspergillus. During drying coffee beans was contaminated by 4 species namely Aspergillus terreus, A. repens, Cladosporium sp. Mucor sp. Whereas during storage was contaminated by 4 spesies namely A. chevalieri, A. niger, Penicillium citrinum, Rhizopus sp. A total of 11 strains A. flavus were isolated consisted of 3 strains isolated from soil at plantation area, 7 strains from coffee beans during drying and 1 strain during storage. Based on the toxigenicity ten strains A. flavus were toxigen (aflatoxin producers), 3 strains were isolated from from soil plantation, 6 strains from coffee beans during drying and 1 strains isolated from coffee beans during storage. Semi-quantitative aflatoxin analysis by thin layer chromatography showed that A. flavus isolated from soil at plantation area produced the highest level of aflatoxin 42.4 μg/kg.en_US
