Pengaruh Pemberian Kombinasi Ekstrak Taoge dan Kinetin terhadap Pertumbuhan Tunas Anggrek Dendrobium sp. pada Media MS

Abstract

Penelitian mengenai Pengaruh Pemberian Kombinasi Ekstrak Taoge dan Kinetin Terhadap Pertumbuhan Tunas Anggrek Dendrobium sp. pada Media MS telah dilaksanakan dari bulan April 2021 sampai bulan September 2021 di Laboratorium Kultur Jaringan UPT. Benih Induk Hortikultura Gedung Johor Medan, Sumatera Utara. Penelitian ini bertujuan untuk mendapatkan kombinasi konsentrasi ekstrak taoge dan kinetin terbaik dalam pertumbuhan tunas anggrek Dendrobium sp. secara in vitro. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) faktorial dua faktor dengan empat taraf ekstrak taoge (0, 5, 10, dan 15 %) dan empat taraf kinetin (0, 0,5, 1, dan 1,5 ppm). Hasil uji statistik menunjukkan bahwa kombinasi konsentrasi ekstrak taoge dan kinetin berpengaruh nyata terhadap pembentukan jumlah daun, tinggi planlet, jumlah akar, panjang akar, berat basah dan berat kering (p ˂ 0,05). Kombinasi perlakuan terbaik didapatkan dari konsentrasi ekstrak taoge 10% dan kinetin 1 ppm (N2K2) menghasilkan rata-rata jumlah tunas sebanyak 2,33 tunas, jumlah daun sebanyak 5,67 helai, dan jumlah akar sebanyak 4,11.

Research on the Effect of the Combination of Sprout Extract and Kinetin on the Growth Dendrobium sp. Orchid Shoot on MS Medium has been carried out from April 2021 to September 2021 at the UPT Tissue Culture Laboratory, Horticulture Seed Center, Gedung Johor, Medan, North Sumatera. This research aimed to determine the best combination concentration of bean sprout extract and kinetin in the growth of Dendrobium sp. orchid shoot in vitro. This research used a two factor factorial Completely Randomized Design (CRD) with four levels of bean sprout extract (0, 5, 10, and 15%) and four levels of kinetin (0, 0,5, 1, and 1,5 ppm). The statistic result showed that the combination of bean sprout extract concentration and kinetin significantly affected the formation of leaf number, plantlet height, number of roots, root length, wet weight and dry weight (p˂0.05). The best treatment combination was obtained from a concentration 0f 10% bean sprout extract and 1s ppm kinetin (N2K2) resulting in an average number of shoots of 2,33 shoots, number of leaves of 5,67 strands, and the number of roots of 4,11.
