dc.description.abstract | Application of pesticides especially in developing countries, on the agricultural sector has become a common thing. Pesticides widely used is a synthetic pesticide that has a negative impact on the environment, such as causing pollution and the accumulation of pesticide residues, which finally goes into the food chain. Research to determine the viability of the immobilized biosurfactant producing bacteria and its ability to degrade pesticides has been carried out. This study used Pseudomonas aeruginosa and Pseudomonas aeruginosa Strain M 111, collections of the Laboratory of Microbiology, Faculty Mathematics and Natural Sciences, University of Sumatera Utara. Those bacteria were immobilized on two different types of immobilizing materials, alginate and polyurethane foam (PUF). The result showed that both immobilizing materials have higt effectiveness (97.97% in alginate and 93.24% in PUF). Bacterial viability analysis showed that the bacteria immobilized with alginate was better than that of the polyurethane during 15 days of incubation at ambient temperature. However, for application on the Busnel Hass Broth (BHB) containing carbofuran as the sole carbon source, the bacteria immobilized using PUF have better viability than that in alginate. Macroscopic observation showed that alginate capsules tended to disolve during 15-day incubation period. Analysis of carbofuran residue using Hight Performance Liquid Chromatography (HPLC) indicated that both isolates immobilized with alginate have able to degrade carbofuran completely (initial concentration of 41,36 ppm) during 5 days of incubation and bacteria immobilized with PUF were able to degrade carbofuran completely during 15 days of incubation. | en_US |
dc.description.abstract | Penggunaan pestisida di negara berkembang khusunya pada sektor pertanian sudah menjadi suatu hal yang lazim. Pestisida yang umum digunakan merupakan pestisida sintetis yang memiliki dampak negatif terhadap lingkungan, seperti pencemaran dan akumulasi dari residu pestisida yang pada akhirnya masuk ke dalam rantai makanan. Penelitian untuk mengetahui viabilitas bakteri penghasil biosurfaktan terimobilisasi dan kemampuannya dalam mendegradasi pestisida telah dilakukan. Pada penelitian ini digunakan bakteri Pseudomonas aeruginosa dan Pseudomonas aeruginosa Strain M 111 koleksi Laboratorium Mikrobiologi, FMIPA Universitas Sumatera. Bakteri diimobilisasi di dua jenis bahan penyalut yaitu alginat dan polyurethane foam (PUF). Efektifitas bahan penyalut dalam memerangkap bakteri terbilang tinggi tinggi masing-masing yaitu 97,97% untuk alginat dan 93,24% untuk PUF. Analisis viabilitas bakteri menunjukkan bahwa bakteri yang diimobilisasi dengan alginat lebih baik dibandingkan dengan polyurethane selama 15 hari masa inkubasi pada suhu ambient. Namun untuk pengaplikasian pada media uji, bakteri yang diimobilisasi menggunakan PUF memilki viabilitas yang lebih baik dibandingkan dengan alginat. Pengamatan makroskopik menunjukkan bahwa kapsul alginat cenderung mengalami peleburan selama 15 hari masa inkubasi. Analisis kemampuan degradasi pestisida oleh bakteri pada media Busnel Hass Broth yang mengandung karbofuran menggunakan Hight Performance Liquid Chromatography . Hasil menunjukkan bahwa kedua isolat yang diimobilisasi dengan alginat mampu mendegradasi 100% karbofuran (konsentrasi awal 41,36 ppm) selama 5 hari masa inkubasi dan bakteri yang diimobilisasi pada PUF mampu mendegradasi 100% karbofuran dalam 15 hari. | en_US |
