    Perkecambahan Biji Tanaman Biwa (Eriobotrya japonica Lindl.) dengan Penambahan GA3 dan Kinetin Secara In Vitro

    Abstract
    This study has been done from April to August 2016 at the Physiology and Plant Tissue Culture Laboratory of Biology Department, Faculty of Mathematics and Natural Science, University of Sumatera Utara, Medan. The study aimed to determine the best concentration of GA3, kinetin or the combination on biwa seed germination. The experiment was carried out using a completely randomized design with two factors. The first factor are three levels of GA3 concentrations; 0, 250 and 500 ppm. The second factor are four levels of kinetin concentrations; 0, 4, 8 and 12 ppm. The results showed that there is significant effect of GA3, kinetin and the combination application on emergenced of germination and fresh weight of root with the best concentrations are 500 ppm GA3 + 8 ppm kinetin and 250 ppm GA3 + 4 ppm kinetin. The application did not significantly affected on percentage of germination, height of seedling, amount of shoot, amount of leaf, fresh and dry weight of shoot. GA3 without kinetin application significantly affected on dry weight of root with the best concentration is 500 ppm GA3.
     
    Penelitian ini dilakukan dari bulan April sampai bulan Agustus 2016 di Laboratorium Fisiologi dan Kultur Jaringan Tumbuhan Departemen Biologi Fakultas Matematika dan Ilmu Pengetahuan Alam, Universitas Sumatera Utara. Tujuan penelitian ini untuk mendapatkan konsentrasi terbaik dari GA3, kinetin atau kombinasi keduanya terhadap perkecambahan dan pertumbuhan biji tanaman biwa. Rancangan yang digunakan adalah rancangan acak lengkap (RAL) faktorial dua faktor dengan tiga taraf GA3, yaitu 0 ppm; 250 ppm; 500 ppm dan empat taraf kinetin, yaitu 0 ppm; 4 ppm; 8 ppm; 12; ppm. Konsentrasi GA3, kinetin dan interaksi keduanya memperlihatkan pengaruh nyata terhadap waktu muncul kecambah dan berat basah akar dengan perlakuan terbaik berturut-turut adalah GA3 500 ppm + kinetin 8 ppm dan GA3 250 ppm + kinetin 4 ppm. Pengaruh yang tidak nyata ditunjukkan terhadap persentase daya berkecambah, tinggi kecambah, jumlah tunas, jumlah daun, berat basah tajuk dan berat kering tajuk. Pemberian GA3 secara tunggal memperlihatkan pengaruh nyata terhadap nilai berat kering akar dengan perlakuan terbaik adalah GA3 500 ppm.
