    Kemampuan Aspergillus Niger dan Saccharomyces Cerevisiae serta Metabolit yang Dihasilkan dalam Menghambat Pertumbuhan dan Produksi Aflatoksin Galur-Galur Aspergillus Flavus

    Abstract
    The contamination of Aspergillus flavus on agricultural commodities cause physical damage and produce aflatoxins that harmful to humans. This study aimed to determine strains of Aspergillus niger and Saccharomyces cerevisiae and their metabolites to inhibit the growth and production of aflatoxins of Aspergillus flavus strains. Fungi used in this study were A. niger AN01, AN02, AN03 and AN04, S. cerevisiae, toxigenic A. flavus (TKA6, KAF11, TK02, TK03), and non-toxic A. flavus (KPJ7, KAF7). Parameters observed were antagonistic mechanism using dual culture method, secondary metabolite production (citric acid, acetic acid) and the effect of the organic acids on the growth and production of aflatoxin by toxigenic A. flavus. Results showed that four strains of A. niger and S. cerevisiae against toxigenic and non-toxigenicc A. flavus with the percentage inhibition of 40.74 to 75%. Secondary metabolites produced by A. niger (citric acid) and S. cerevisiae (acetic acid) were potential to inhibit the growth of toxigenic A. flavus and their aflatoxin production. Two strains of A. niger (AN01 and AN04) were the highest citric acid production (0,173% and 0,091%), whereas S. cerevisiae produce 0,046% acetic acid. The organic acids have effect on the growth and production aflatoxins. Semi-quantitative aflatoxin production using TLC showed a decrease in aflatoxin production up to 68,3 μ/kg.
     
    Serangan cendawan Aspergillus flavus pada komoditas pertanian menyebabkan kerusakan fisik serta menghasilkan aflatoksin yang berbahaya bagi manusia. Penelitian ini bertujuan mengetahui kemampuan beberapa galur Aspergillus niger dan Saccharomyces cerevisiae serta metabolit sekunder yang dihasilkan dalam menghambat pertumbuhan dan produksi aflatoksin galur-galur Aspergillus flavus. Fungi yang digunakan dalam penelitian ini adalah galur A. niger AN01, AN02, AN03, AN04, S. cerevisiae dan galur A. flavus toksigen TKA6, KAF11, TK02, TK03, serta galur A. flavus non-toksigen KPJ7, dan KAF7. Kemampuan antagonis dilakukan dengan metode dual culture, analisis metabolit sekunder oleh A. niger (asam sitrat) dan S. cerevisiae (asam asetat) serta pengaruhnya terhadap pertumbuhan dan produksi aflatoksin secara kualitatif dan semi-kuantitatif. Uji antagonis 4 galur A. niger dan S. cerevisiae terhadap cendawan A. flavus toksigen dan non toksigen menunjukkan kemampuannya berkompetisi dengan persentase hambatan 40,74 hingga 75%. Aspergillus niger dan S. cerevisiae berpotensi menghambat pertumbuhan A. flavus toksigen dan produksi aflatoksin. Dua galur A. niger yaitu AN01 dan AN04 memiliki persentase kandungan asam sitrat tertinggi masing-masing 0,173 dan 0,091%. Saccharomyces cerevisiae menghasilkan asam asetat 0,046%. Analisis produksi aflatoksin secara semi-kuantitatif menggunakan KLT menunjukkan efek asam organik menunjukkan penurunan produksi aflatoksin hingga 68,3 μg/kg.
