    Korelasi Polimorfisme VEGF -2578 C>A terhadap Lesi Prakeganasan Gaster Pada Pasien Gastritis Helicobacter Pylori

    Abstract
    Gastric cancer is one of the most common causes of cancer-related death worldwide. Many epidemiological studies show several risk factors associated with the development of gastric cancer, including gastritis. The most common cause of gastritis is H. pylori infection. In the mucosa that was infected with H. pylori, there was a significant increase in VEGF levels. Several SNPs are suspected to be involved as the etiology of malignancy, one of which is the VEGF- 2578C> A polymorphism. Objective: The aim of this study is to evaluate the involvement of the VEGF -2578C>A gene polymorphism to gastric premalignant conditions in susceptible individual such as gastritis patients. Method: A cross sectional study was conducted in a field with 59 gastritis H.pylori patients. Endoscopic examination. Endoscopic examination is performed to assess the gastric mucosa. H. pylori status was determined by examining Urea breath test (UBT) and Campylobacter Like Organism (CLO) test followed by histopathological studies that evaluated any changes to premalignant lesions such as chronic atrophic gastritis, intestinal metaplasia, intestinal dysplasia. Genotype analysis was performed using real-time polymerase chain reaction (RT-PCR). Result. Fifty nine people patients with gastritis H. pylori 35.8% of patients showed premalignant lesions. The majority of premalignant lesions had genotype CC (37.9%, n = 11), but there was no significant relationship between VEGF -2578 C> A polymorphisms and the incidence of premalignant gastric lesions. Conclusion. Based on this study, the VEGF -2578C> A polymorphism was not associated with gastric premalignant lesions.
     
    Kanker Gaster merupakan salah satu penyebab tersering kematian terkait kanker di seluruh dunia. Banyak penetian epidemiologi menunjukan beberapa faktor resiko yang berhubungan dengan perkembangan dari kanker gaster, termasuk gastritis . Penyebab tersering dari gastritis adalah infeksi H. pylori . Pada mukosa yang sudah terinfeksi H.pylori terjadi peningkatan dari kadar VEGF secara signifikan. Beberapa SNP diduga terlibat sebagai etiologi keganasan, salah satunya adalah polimorfisme VEGF-2578C>A Tujuan: Tujuan dari penelitian ini adalah untuk mengevaluasi keterlibatan polimorfisme gen VEGF - 2578C> A terhadap kondisi prakeganasan gaster pada pasien dengan gastritis H. pylori Studi cross sectional dilakukan di medan dengan 59 pasien gastritis H. Pylori. Pemeriksaan endoskopi. Pemeriksaan endoskopi dilakukan untuk menilai mukosa lambung. Status H.pylori ditentukan dengan pemeriksaan Urea breath Test (UBT) dan test Campylobacter Like Organism(CLO) diikuti dengan studi histopatologis yang mengevaluasi adanya perubahan menjadi lesi pra keganasan seperti gastritis atrofi kronis , metaplasia usus, displasia usus. Analisis genotipe dilakukan dengan menggunakan polymerase chain reaction (RT-PCR) secara real-time. Hasil. Dari 59 pasien pasien gastritis H.pylori , 35.8 % pasien menunjukan lesi prakeganasan. Mayoritas lesi pramaligna memiliki genotip CC (37.9%, n = 11), namun tidak ada hubungan yang bermakna antara polimorfisme VEGF -2578 C> A dengan kejadian lesi prakeganasan gaster. Kesimpulan. Berdasarkan penelitian ini, polimorfisme VEGF -2578C> A tidak berhubungan dengan lesi prakeganasan gaster pada pasien gastritis H. pylori
