Pengaruh Ekstrak Daun Puguntano (Picria fel-terrae (Lour.) Merr.) terhadap Histomorfometri Aorta Jantung Pada Tikus Galur Wistar (Rattus norvegicus) Model Obesitas dengan Benign Prostat Hyperplasia

Abstract

Obesity is a condition in which there is an accumulation of excess fat in the body. The use of puguntano leaf extract has bioactive compounds that can reduce the level of damage to the aorta. This study aims to determine the effect of leaf extract of puguntano (Picria fel-terrae (Lour.) Merr.) on the histomorphometry of the heart of obese rats (Rattus novergicus) with benign prostatic hyperplasia. This study used a completely randomized design (CRD) with 5 groups and 5 replications. The group in this study consisted of a negative control group (without treatment), positive control (obese mice with BPH and testosterone injection), the first treatment (obese mice with BPH and testosterone injection and given standard drug finesteride), the second treatment (obese mice with BPH and injected with testosterone and given puguntano leaf extract 100 mg/KgBW), the third treatment (obese rats with BPH and injected with testosterone and given puguntano leaf extract 200 mg/KgBW). Obese rats were identified by calculating Body Mass Index (BMI). Aortic organ preparations were made by paraffin method and Hematoxylin Eosin (HE) staining. The results showed that the puguntano leaf extract treatment of 200 mg/KgBW had an effect on improving the histomorphometry of the rat heart which resulted in heart weight, heart volume, aortic wall thickness, aortic diameter, respectively, with an average of 0.83; 1.16; 75.76; 97,42 and reduce the percentage of atherosclerosis frequency to 20%.

Obesitas merupakan suatu keadaan dimana terjadi penumpukan lemak berlebih didalam tubuh. Penggunaan ekstak daun puguntano memiliki senyawa bioaktif yang mampu menurunkan tingkat kerusakan pada aorta. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak daun puguntano (Picria fel-terrae (Lour.) Merr.) terhadap gambaran histomorfometri aorta jantung tikus (Rattus novergicus) obesitas dengan benign prostat hyperplasia. Penelitian ini menggunakan rancangan acak lengkap (RAL) dengan 5 kelompok dan 5 ulangan. Kelompok dalam penelitian terdiri dari kelompok kontrol negative (tanpa perlakuan), kontrol positif (tikus obesitas dengan bph dan diinjeksi testosterone), perlakuan pertama (tikus obesitas dengan bph dan diinjeksi testosterone dan diberi obat standar finesteride), perlakuan kedua (tikus obesitas dengan bph dan diinjeksi testosterone dan diberi ekstrak daun puguntano 100 mg/KgBB), perlakuan ketiga (tikus obesitas dengan bph dan diinjeksi testosterone dan diberi ekstrak daun puguntano 200 mg/KgBB). Tikus obesitas diketahui dengan melakukan perhitungan Indeks Massa Tubuh (IMT). Preparat organ aorta dibuat dengan metode paraffin dan pewarnaan Hematoxylin Eosin (HE). Hasil penelitian menunjukkan bahwa perlakuan esktrak daun puguntano 200 mg/KgBB berpengaruh untuk memperbaiki histomorfometri aorta jantung tikus yang menghasilkan berturutturut berat jantung, volume jantung, ketebalan dinding aorta, diameter aorta dengan rata-rata sebanyak 0,83; 1,16; 75,76; 97,42 dan menurunkan persentase frekuensi aterosklerosis menjadi 20%.
