Uji Toksisitas Ekstrak Daun Pirdot (Saurauia vulcani Korth.) terhadap Gambaran Histologis Hepar Mencit (Mus musculus L.)

Abstract

The research on the toxicity of pirdot leaf extract (Saurauia vulcani Korth.) on the histological feature of the liver pregnant mice (Mus musculus L.) was carried out from June to December 2021 at the Animal Physiology Laboratory, Animal House Laboratory, Organic Chemistry Laboratory of the USU Faculty Mathematic and Natural Science and Anatomy and the Phatology Laboratory of the hospital Adam Malik. The pirdot plant is one of the local plants of North Sumatra that is used by the community as herbal ingredients for various diseases such as diabetes. The utilization of pirdot leaf extract for diabetes therapy needs futher studies such as teratology studies. The aim of study was to determine the toxicity of pirdot leaf extract on the liver of pregnant mice as part of the maternal effect teratology test. This study used a completely randomized design (CRD) method using 50 pregnant mice which were divided into 5 group treatments with 10 replications on each. Two treatments were negative control (K-), without any treatment and positive control (K+) given palmitate vitamin A of 200000 IU/kgBW, and 3 treatments were given pirdot leaf extract at doses of 300 mg/kgBW, 600 mg/kgBW and 900 mg/kgBW, respectively. The pirdot leaf were given using gavage on day 0 to day 16 of pregnancy. Then pregnant mice’s body weight was weighed every day. The results showed that the administration of pirdot leaf extract at a dose of 600 mg/kgBW significantly reduced body weight on days 2, 4 and 6 of the time -pregnancy, also significantly reduced the liver weight of pregnant mice at doses of 300 and 600 mg/kgBW. It also affect significantly to the liver histopathological changes in pregnant mice. However, the administration of pirdot leaf extract in pregnant mice did not significantly affect the liver morphology in pregnant mice.

Penelitian uji toksisitas ekstrak daun pirdot (Saurauia vulcani Korth.) terhadap gambaran histologis hepar mencit (Mus musculus L.) bunting telah dilakukan pada bulan Juni sampai Desember 2021 di laboratorium Fisiologi Hewan, laboratorium Rumah Hewan Percobaan, laboratorium Kimia Organik FMIPA USU dan laboratorium Patologi Anatomi Rumah Sakit Adam Malik. Tumbuhan pirdot merupakan salah satu tumbuhan lokal Sumatera Utara yang digunakan masyarakat sebagai bahan herbal untuk berbagai penyakit seperti diabetes. Pengembangan ekstrak daun pirdot untuk terapi diabetes masih membutuhkan kajian keamanan termasuk kajian teratologi. Penelitian ini bertujuan untuk mengetahui toksisitas ekstrak daun pirdot pada hepar mencit bunting sebagai bagian dari uji teratologi efek maternal. Penelitian ini menggunakan metode Rancangan Acak Lengkap (RAL) menggunakan 50 ekor mencit bunting yang dibagi ke dalam 5 kelompok perlakuan masing-masing 10 ulangan. Dua kelompok perlakuan merupakan kontrol negatif (K-) tanpa perlakuan dan kontrol positif (K+) diberi vitamin A palmitate 200.000 IU/kgBB, dan 3 kelompok perlakuan diberi ekstrak daun pirdot dosis 300 mg/kgBB, 600 mg/kgBB, dan 900 mg/kgBB. Perlakuan diberikan secara oral dengan cara pencekokan menggunakan gavage pada hari ke-0 hingga hari ke-16 kebuntingan. Kemudian ditimbang berat badan mencit bunting setiap hari. Hasil penelitian menunjukkan pemberian ekstrak daun pirdot dosis 600 mg/kgBB pada mencit bunting memengaruhi penurunan berat badan secara signifikan pada hari ke- 2, 4 dan 6 kebuntingan, dan juga signifikan menurunkan berat hepar mencit bunting pada dosis 300 dan 600 mg/kgBB, serta signifikan memengaruhi perubahan histopatologi hepar mencit bunting. Namun, pada pemberian ekstrak daun pirdot pada mencit bunting tidak memengaruhi secara signifikan morfologi hepar mencit bunting
