Hubungan Polimorfisme Gen ACTN-3 Terhadap Daya Ledak Otot (Muscular Power) pada Siswa Sekolah Sepakbola Kota Medan

Abstract

The ACTN-3 gene is a gene that The ACTN-3 gene is a gene that encodes a sarcoma protein almost entirely expressed in fast glicolytic muscle and plays an important role in the speed and contraction of high muscle strength or high-blown contraction generator, while the phenotype of polymorphism in the ACTN-3 gene (R/X) tend to have better endurance. Explosive power is the ability of the muscle to overcome the load resistance with high strength and speed in a complete movement. Explosive power is an important component in football. The purpose of this study is to analyze the relationship between ACTN-3 gene polymorphism related to muscular power explosive power. The research design used in this research is analytic descriptive by using cross sectional approach. The subjects of this study were soccer school students who were in Medan amounted to 33 people, 11-14 years old, BMI as many as 5 people (15.2%) and a normal BMI of 28 people (84.8%). Muscle power muscularity was measured using a standing broad jump test and the ACTN-3 gene polymorphism was identified using PCR-RFLP from the buccal cell sample of the study subjects. Fisher Exact test was used to investigate the relationship between ACTN-3 gene polymorphism with explosive muscle limb power. The results of the study suggest that there is a significant relationship between ACTN-3 gene polymorphism and explosive muscle limb in Medan soccer school students (p = 0.00)

Gen ACTN-3 merupakan gen yang mengkode sebuah protein sarkomer yang hampir secara keseluruhan diekspresikan dalam serat otot cepat (fast glicolytic) dan berperan penting dalam kecepatan dan kontraksi kekuatan otot yang penuh atau pembangkit ledakan kontraksi yang penuh, sementara fenotip polimorfisme pada gen ACTN-3 (R/X) cenderung memiliki daya tahan yang lebih baik. Daya ledak (power) adalah kemampuan otot untuk mengatasi tahanan beban dengan kekuatan dan kecepatan tinggi dalam suatu gerakan yang utuh. Daya ledak merupakan komponen penting dalam sepakbola. Tujuan penelitian ini yaitu menganalisis hubungan antara polimorfisme gen ACTN-3 terhadap daya ledak otot tungkai (muscular power). Desain penelitian yang digunakan pada penelitian ini adalah deskriptif analitik dengan menggunakan pendekatan cross sectional. Subjek penelitian ini merupakan siswa sekolah sepakbola yang di kota Medan berjumlah 33 orang, usia 11 – 14 tahun, IMT kurus sebanyak 5 orang (15,2%) dan IMT normal sebanyak 28 orang (84,8%). Daya ledak otot tungkai (muscular power) diukur menggunakan tes standing broad jump dan polimorfisme gen ACTN-3 diidentifikasi menggunakan PCR-RFLP dari sampel sel bukal subjek penelitian. Uji Fisher Exact digunakan untuk mengetahui hubungan antara polimorfisme gen ACTN-3 dengan daya ledak otot tungkai. Hasil studi menyatakan bahwa ada hubungan signifikan antara polimorfisme gen ACTN-3 dengan daya ledak otot tungkai pada siswa sekolah sepakbola kota Medan (p=0,00).
