    Hubungan Polimorfisme Genetik CYP1A1 Ile462Val terhadap Kejadian Kanker Paru di RSUP H. Adam Malik Medan

    Abstract
    Lung cancer is malignancy that ranks first in male and female in Indonesia who are associated with smoking. Genetic polymorphism plays an important role in carcinogenesis. Polymorphism of CYPIAI Ile462val proved to convert benzo (a) pyrene in cigarettes into an active form of BP-7,8-dihydrodiol-9, 10-epoxide which is a potent carcinogen. The aims of this study is to analyze the relationship between genetic polymorphism CYPIAI Ile462val with lung cancer incidence Method This was a case control study involving 80 research subjects through consecutive sampling technique from Adam Malik General Hospital in Medan, Indonesia. The examination of the CYPIA! gene was conducted with Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP). Data were analysed with Conditional Logistic Regression Test using Epi-Info 7 software. Result The allele frequencies of wildtype AA, AG, and GG found were 62.5%, 25%, and 12,5%, respectively. There was no significant relationship between CYPIAI allele with lung cancer incidence (p-0.09). Conclusion There was no association between CYPIAI genetic polymorphism with hing cancer susceptibility and requires further study.
     
    Pendahuluan Kanker paru merupakan suatu keganasan yang menduduki peringkat pertama pada laki-laki dan perempuan di Indonesia yang berhubungan dengan merokok. Polimorfisme genetik mempunyai peran dalam karsinogenesis. Polimorfisme CYPIAI Ile462Vl terbukti dapat mengubah benzo (a) pyrene dalam rokok menjadi bentuk aktif dari BP-7,8-dihydrodiol-9,10-epoxide yang berpotensi menjadi karsinogen. Penelitian ini bertujuan untuk menganaltsis hubungan antafa polimorfisme genetik CYPlAl Ile462val dengan insidensi kanker paru. Metode Penelitian ini menggunakan pendekatan studi analitik kasus kontrol yang melibatkan 80 subjek penelitian dengan teknik konsekutif sampling. Penelitian ini dilakukan di Rumah Sakit Umum Pusat Haji Adam Malik, Indonesia. Pemeriksaan gen CYPlAl dilakukan dengan Polymerase Chain ReactionRestriction Fragent Length Polymorphism (PCR-RFLP). Data dianalisis dengan Conditional Logistic Regression Test menggunakan Epi-Jnfo 7 software. Basil Frekuensi alel dari wildtype AA, AG dan GG yang ditemukan adalah 62,5%, 25% dan 12,5% secara berurutan. Tidak terdapat hubungan yang signifikan antara alel CYPlAl dengan insidensi kanker paru (p=0,09). Kesimpulan Tidak terdapat hubungan antara polimorfisme genetik CYPlAl dengan kejadian kanker paru dan membutuhkan penelitian yang lebih lanjut.
