    Isolasi dan Karakterisasi Kitinase Isolat Bakteri dari Usus Halus Kadal Tanah (Mabouya multifasciata) dan Tokek (Gekko gecko)

    Abstract
    A study on isolation and characterization of partial purification of chitinase enzyme isolated from the reptile small intestine have been done. The aims of this study are to identify chitinolytic bacteria, measurement of chitinase activity and chitinase enzyme characterization. Isolation was conducted on Minimum Salt Chitin Medium (MSCM) and incubated at 31 0C. Chitinolytic index was measured by using minimum salt chitin medium incubated for 5 days. Two isolates with the highest chitinolytic index were selected measured for their enzyme activity by Spindler method. Partial purification of enzyme was began by precipitation using amonium sulphate in different stages of different concentrations and precipitate was dialized. Effect of pH and temperature was done to know the characteristics of each enzyme produced . The highest crude extract spesific activity was obtained from KD5 isolate of 0.013 U/mg and LK12 of 0.0107 U/mg on the sixth day. Chitinase enzyme from KD5 and LK12 were optimally precipitated at 60% and 50% concentrations of amonium sulphate, with specific activity values of 0.0647 U/mg and 0.0585 U/mg. Post-dialysis specific activity also showed significant increase on KD5 and LK12 isolates of 0.1527 U/mg and 0.0802 U/mg. Based on 16S rRNA identification KD5 isolate have 88% similarity with Enterobacter tabaci starin YIM Hb-3 and LK12 Isolate have 91% similarity with Stenothrophomonas maltophilia strain ATCC 19861.
     
    Penelitian mengenai isolasi dan karakterisasi parsial enzim kitinase yang diisolasi dari saluran pencernaan reptil telah dilaksanakan. Penelitian ini bertujuan untuk mengidendifikasi bakteri kitinolitik, uji aktivitas kitinase dan karakterisasi enzim kitinase. Isolasi dilakukan pada media spesifik yaitu media MGMK dan diinkubasi pada suhu 31 oC. Uji indeks kitinolitik dilakukan juga dengan menggunakan media MGMK dan diinkubasi selama 5 hari. Dua isolat terpilih dengan nilai indeks kitinolitik tertinggi kemudian diukur aktivitas enzim kitinasenya dengan menggunakan metode Spindler. Presipitasi enzim parsial dilakukan dengan menambahkan garam amonium sulfat secara bertingkat dengan konsentrasi yang berbeda beda untuk selanjutnya didialisis. Pengaruh pH dan suhu dilakukan untuk mengetahui karakteristik masing masing enzim yang dihasilkan isolat bakteri. Aktivitas spesifik kitinase ekstrak kasar tertinggi diperoleh dari isolat KD5 sebesar 0,013 U/mg dan isolat LK12 sebesar 0,0107 U/mg dengan masing-masing puncak produksi pada hari keenam. Enzim kitinase dari isolat KD5 dan LK12 dipresipitasi optimal pada konsentrasi garam 60% dan 50%, dengan nilai aktivitas spesifik sebesar 0,0647 U/mg dan 0,0585 U/mg. Aktivitas spesifik pasca dialisis juga menunjukkan peningkatan yang signifikan pada isolat bakteri KD5 dan LK12 yaitu sebesar 0,1527 U/mg dan 0,0802 U/mg. Berdasarkan Identifikasi gen 16s rRNA isolat KD5 memiliki kemiripan 88% dengan Enterobacter tabaci strain YIM Hb-3 dan isolat LK12 memiliki kemiripan 91% dengan Stenothrophomonas maltophilia strain ATCC 19861.
