    Pengaruh Curcuminoid terhadap Ekspresi Calcineurin (Cn) dan Nuclear Factor of Activated T-Cells Cytoplasmic 1 (NFATc1) pada Kejadian Apoptosis Fibroblas Koklea Rattus Norvegicus yang Diberi Pajanan Bising

    Abstract
    Background: The structural changes underlying noise-induced hearing loss (NIHL) include loss of the sensory hair cells, damage to their stereocilia and supporting tissues within the cochlear lateral wall. Objective: To demonstrate curcuminoid as safe and effective therapeutic agent in the prevention and treatment for fibroblasts damage within the cochlear supporting tissues and lateral wall through cell death pathway. Methods: The samples were 24 Rattus norvegicus divided into 4 groups. Group 1: control group; group 2: noise (+); group 3: noise (+), 50 mglday curcuminoid (+;; group 4: noise (+), 100 mg/day curcuminoid (+). The noise exposure dose given was 100 dB SPL, frequency range is 1 to 10 kHz for 2 hours in 2 weeks and the curcuminoid were administered orally in 2 weeks. This study used curcuminoid derived from Curcuma longa Linnaeus (Turmeric) with curcuminoid content levels I28.1!1.01% w/w compared to standard. All samples were examined for the expression of Calcineurin (Cn) dan Nuclear Factor of Activated T-Cells Cytoplasmic 1 (NFATcl ) by immunohistochemistry method, as well as the number of cochlear fibroblasts apoptosis by TUNEL assay. Results: The results obtained showed significant differences for the expressions of Cn (p . O.OS) in all groups, significant differences for the expressions of NFATcI (p < 0.05) in all groups, except in group 1 and 4, and significant differences for the apoptotic index (p . O.OS) in ail groups. Positive correlation between the expression of Cn and NFAI-c1 with cochlear fibroblasts apoptosis in all groups. Curcuminoid proved to be potentially effective in the prevention and treatment for fibroblasts damage within the cochlear supporting tissues and lateral wall regarding the decreased expression of Cn, NFATcI and apoptotic index of cochlear fibroblasts
     
    Latar belakang: Perubahan struktural yang mendasari gangguan pendengaran akibat bising (GPAB) termasuk hilangnya sel-sel rambut sensorik, kerusakan stereosilia dan jaringan-jaringan penyokong dinding lateral koklea. Tujuan penelitian: l\4embuktikan curcunthroid sebagai agen terapeutik yang efektif dan aman dalam mencegah dan mengatasi kerusakan fibroblas jaringan-jaringan penyokong dan dinding lateral koklea melalui jalur kematian sel. Metode penelitian: Sampel yang digunakan dalam penelitian ini adalah 24 Raffus noruegicus yang dibagi menjadi 4 kelompok. Kelompok 1. kontrol, kelompok 2: bising (+), kelompok 3: bising (+) curcuntinoid 50 mg/hari (+), kelompok 4: bising (+) curcuminoid 100 mg/hari (+;. Dosis bising yang diberikan adalah 100 dB SPL dengan frekuensi 1 s/d 10 kHz selama 2 jam, selama 2 minggu, dan curcuntinoid diberikan secara oral selama 2 minggu. Penelitian ini menggunakan curcuminoid yang berasal dari Curcuma longa L, (Turmeric) dengan kadar curcuntinoid [28.1t1 0]o/o b/b dibanding standar. Pada semua sampel dilakukan pemeriksaan ekspiesi- Cglcineurin (Cn) dan Nuclear Factor of Activated T-Cells Cytoplasmie'7 (NFATcl) dengan metode imunohistokimia, serta jumlah fibroblas dinding lateral koklea yang mengalami apoptosis dengan TUNEL assay. Hasi!: Hasil pemeriksaan diperoleh terdapat perbedaan ekspresi Cn yang bermakna (p < 0.05) untuk semua kelompok perlakuan, ekspresi NFATcI yang bermakna (p . 0.05) untuk semua kelompok perlakuan, kecuali antqra kelompok 1 dan 4, dan jumlah sel yang mengalami apoptosis yang beFmakna,(p < 0 05) untuk-semua_kelompok perlakuan. Korelasi positif diantara ekspresi Cn dan NFATcI dengan apoptosis fibroblas koklea. Curcuminoid terbukti efektif berpotensi mencegah dan mengatasi kerusakan fibroblas jaringan-jaringan penyokong dan dinding lateral koklea berdasarkan penurunan ekspresi Cn, NFATcI dan jumlah sel yang mengalami apoptosis
