dc.description.abstractIntroduction Tetracycline have been widely used as periodontal supportive treatment since it has broad spectrum, bone tissue penetration and MMP and collagenase inhibition native. Chitosan hydrogel based tetracycline are known to have strong antibacterial effect, but the cytotoxic effect on fibroblast are yet to be studied. Objective To obtain the in vitro cytotoxicity effect of chitosan hydrogel based tetracycline on fibroblast as supportive periodontal treatment. Material and Method The 0.5%,0.7% and 1% chitosan hydrogel based tetracycline, chitosan hydrogel without tetracycline are made by dissolving chitosan powder with citric acid, followed by tetracycline powder. 3T3 fibroblast cells were cultured in well-microplate containing RPMI-1640 media inside incubator. Viability assay were carried out with MTT-assay method and repeated for 5 times. Degradation of color were read with microplate reader Result The mean viability of fibroblast applied with chitosan hydrogel based tetracycline0.5%, 0.7%, 1% and chitosan hydrogel are respectively 86.5%, 85.36%, 80.99% and 90.85%. One-way ANOVA test with p<0.05 shows the cell's viability on 1% concentration group significantly differs from the rest. The 0.5% concetration group shows the highest mean of viability. Conclusion Chitosan hydrogel based tetracycline shows low cytotoxic effect on 3T3 fibroblast cell.en_US
dc.description.abstractLatar belakang: Tetrasiklin merupakan obat yang sering digunakan sebagai penunjang perawatan periodontal. Pemberian tetrasiklin secara lokal bertujuan menjaga konsentrasi obat dalam sulkus periodontal serta menghambat kerja MMPs dan enzim kolagenase. Tetrasiklin berbasis hidrogel kitosan diketahui memiliki efek anti bakteri yang baik, namun belum pernah dilakukan penelitian mengenai efek sitotoksiknya terhadap sel fibroblas. Tujuan: Untuk mengetahui sitotoksisitas tetrasiklin berbasis hidrogel kitosan terhadap sel fibroblas sebagai bahan penunjang terapi periodontal secara in vitro. Metode: Pembuatan gel tetrasikin berbasis hidrogel kitosan 0,5%, 0,7%, 1% berbasis hidrogel kitosan dan hidrogel kitosan tanpa tetrasiklin dimulai dengan melarutkan bubuk kitosan dalam larutan asam sitrat diikuti dengan penambahan bubuk tetrasiklin sesuai dengan konsentrasi. Sel fibroblas 3T3 dibiakkan dalam well-microplate yang berisi RPMI-1640 dalam inkubator. Pengujian viabilitas dilakukan dengan metode MTT assay dan diulang sebanyak 5 kali. Perubahan warna dibaca dengan microplate reader. Hasil: Rerata viabilitas sel fibroblas yang diaplikasikan tetrasiklin berbasis hidrogel kitosan 0.5%, 0.7%, 1% dan hidrogel kitosan berturut-turut 86.5%, 85.36%, 80.99% dan 90.85%. Hasil uji statistik ANOVA dengan p≥ 0.05 menunjukkan viabilitas sel fibroblas yang berkontak dengan bahan uji 1% berbeda signifikan dibandingkan konsentrasi lainnya. Gel dengan konsentrasi 0.5% menunjukkan rerata viabilitas paling tinggi. Kesimpulan: Gel tetrasiklin berbasis hidrogel kitosan menunjukkan efek sitotoksik yang rendah terhadap sel fibroblas 3T3.en_US
