Aim:
To Analyze effect of cisplatin on Rattus norvegicus cochlear damage assessed from the VEGF and STAT 1 expression on immunohistochemical examination and otoacoustic emission (OAE) examination.
Methods:
A total of 24 Rattus norvegicus were divided into 4 groups and injected with Cisplatin 8 mg/Kg BW intraperitoneally except for control group. Signal to noise ratio (SNR) values using OAE with frequency 1500-12000 Hz was conducted according to the sample group. Sample was terminated and then the cochlear organ of Corti damage was examined by calculating VEGF and STAT 1 expression on immunohistochemical examination.
Results:
SNR values were decrease in all of the Cisplatin group, especially in the 7th day group. The highest VEGF expression found in the 7th day groups. There was a very strong correlation between SNR values and VEGF also STAT 1 expression in all groups.
Conclusion:
Cisplatin induces VEGF overproduction which leads to cochlear apoptosis, especially hair cells. Cochlear damage due to Cisplatin ototoxicity is shown by a decrease in SNR values accompanied by an increase in VEGF expression in the cochlea.
