Deteksi Virus Gemini pada Tanaman Tomat (Solanum Lycopersicum Mill.) di Kabupaten Simalungun Sumatera Utara dengan Teknik Pcr

Abstract

Geminivirus is a type of virus that attacks tomato plants. The symptoms of the disease caused by the Geminivirus are difficult to distinguish morphologically from other viruses, therefore the Polymerase Chain Reaction (PCR) technique was needed to determine the type of virus in this tomato plants. This method gives a faster and more accurate detection The study aim was to detect the presence of Geminivirus in tomato plants in which showed the disease symptoms at Simalungun District, North Sumatra. The symptoms of viral disease in the tomatoes leaves were observed and collected from 16 villages at Simalungun District. The genomic DNA of tomato leaves which had virus symptoms was isolated and then amplified with Geminivirus specific primers PAL1v 1978-F & PAR1c715-R primers. The results showed that there were 7 symptoms of a viral disease in tomato plants at Simalungun Regency, which were plants stunting, leaf size reducing, leaf edges rolling, leaf chlorosis, curly leaves, mosaics on the leaves, and vein clearing. The most common types of symptoms were found at Saribudolok and Pematang Purba Villages. All samples collected were positive for Gemini Virus in the presence of 1500 bp DNA fragments which were amplified using specific primers
