dc.description.abstractAbstrak Latar belakang : Reseptor vitamin D (RVD) berperan terjadinya kanker, melalui sistem imun serta proliferasi dan diferensiasi sel. RVD terdapat di inti sel guna mengatur ekspresi gen. RVD lebih dari 470 polimorfisme nukleotida tunggal. Polimorfisme Cdx2 (rs 11568820) terletak di regio promoter gen. Cdx2 merupakan faktor transkripsi usus berfungsi meningkatkan penyerapan kalsium.Efek vitamin D dan Kalsium dapat dimediasi oleh RVD. Tujuan : Untuk mengetahui hubungan antara gen RVD Cdx2 dengan kalsium pada penderita kanker payudara Metode : Penelitian analitik dengan desain cross sectional, subjek adalah pasien kanker payudara diRSUP Haji Adam Malik Medan (53 sampel). DNA diisolasi dari darah dengan kit komersial (promega) dan Polimorfisme RVD Cdx2 menggunakan metode allel Spesipik Multiple (ASM) PCR terdapat varian polimorfiseme AA (235 bp, 297 bp) , AG (110 bp, 235 bp, 297 bp) dan GG (110 bp, 297 bp). Pemeriksaan kadar kalsium darah dengan metode o-cresolphelin-complexon dan dianalisa statistik menggunakan uji kruskal wallis dan analisa Hardy Weinberg Equilibrium Hasil : Polimorfisme Cdx2 genotif AA 58,5%, AG 35,8% dan GG 5,7%. Kadar kalsium rata- rata 9,71 + 1,35 mg/d.l dari uji kruskal wallis hasilnya P= 0,07. Nilai HWE diperoleh P = 0,968 ( p > 0,05 ) Kesimpulan : Tidak terdapat hubungan antara gen RVD Cdx2 dengan kalsium pada penderita kanker payudaraen_US
dc.description.abstractBackground: Vitamin D receptors (VDR) involved in cancer pathogenesis, through the immune system, cell proliferation and differentiation. VDR which found in the cell nucleus, responsible in gene expression regulation. More than 470 single nucleotide polymorphisms of VDR have been found. Cdx2 polymorphism (rs 11568820) is located in the gene promoter region. Cdx2 is an intestinal transcription factor that has role in increasing calcium absorption. Vitamin D and Calcium effects can be mediated by VDR. Objective: To determine the relationship between VDR Cdx2 gene polymorphism and calcium level in breast cancer patients Methods: an Analytical study with cross sectional design were conducted. A total of 53 breast cancer patients in RSUP Haji Adam Malik Medan were participated. DNA was isolated from blood with a commercial kit (promega) and VDR Cdx2 Polymorphism were defined using the Multiple Species (ASM) PCR method comprised of AA (235 bp, 297 bp), AG (110 bp, 235 bp, 297 bp) and GG (110 bp, 297 bp) variant. Blood calcium level were measured by the o-cresolphelin-complexon method and analyzed statistically using the Kruskal wallis test and Hardy Weinberg Equilibrium. Results: the AA,AG and GG of Cdx2 polymorphisms were 58.5%, 35.8% and 5.7% respectively. The average calcium level was 9.11 + 1.35 mg / d.l. kruskal wallis test showed that no significant difference were found between genotypes (p = 0.07). This study were in equilibrium of HWE p = 0.968 (p> 0.05) Conclusion: There is no relationship between the RVD Cdx2 gene polymorphism and calcium level in breast cancer patientsen_US
