dc.description.abstractThe Actinomycetes group is known as an antibiotic producer and is still being explored and studied for its ability to inhibit resistant bacteria such as Methicillin-Resistant Staphylococcus aureus (MRSA). This study aims to determine the potency of Actinomycetes from Lau Kawar Lake in inhibiting MRSA bacteria. Actinomycetes were isolated from lake soil and sediment suspended in sodium dodecyl sulfate-yeast extract (SDS-YE) on the Humic acid vitamins Agar (HV-Agar) medium, then The Actinomycetes were characterized macroscopically and microscopically. Primary selection of antibacterial activity of Actinomycetes against MRSA and Staphylococcus aureus ATCC 25923 was carried out by the cross streak method. The potential isolates of MRSA-inhibited by Actinomycetes was fermented and extracted by ethyl acetate solvent and then their inhibition activity were tested againts by the disc diffusion method. The isolates were identified using the Vitek 2 Compact System. The results that 14 Actinomycetes were isolated, namely 10 isolates from soil and 4 isolates from sediment. The macroscopic and microscopic characters showed that all the isolates have irregular and circular colonies with rough and wrinkled surface, undulate, lobate, curled and entire edges, elevated colonies in the form of umbonate, raised, pulvinate. Based on primary selection for the ability of Actinomycetes to inhibit MRSA and S. aureus ATCC 25923 showed that 4 isolates namely TLK06, TLK10, SLK11 and TLK14 potential to inhibit MRSA with inhibition diameters 6 mm to 20 mm. In the antibacterial test of the 4 selected isolates, only one isolate had the largest inhibition zone (21.8 mm), namely TLK06, while the other three isolates ( TLK10, SLK11, TLK14) inhibit less than 21.8 mm and TLK06 was identified as Actinomyces odontolyticus.en_US
