Silkworm cocoon degumming waste contains amino acids in the form of sericin and fibroin which have the potential to stimulate spermatogenesis and to protect sperm from free radicals. This research was carried out from December 2022 to March 2023 at USU's Animal House laboratory, USU's FMIPA natural organic chemistry laboratory, pathology and anatomy laboratory at Adam Malik General Hospital Medan. This study aimed to determine the effect of degumming of silkworm cocoons (Bombyx mori L.) on the morphological and histological improvement of the testes of male mice (Mus musculus L.) that exposed by allethrin. The research design was a completely randomized design (CRD) consisting of 5 treatments; allethrin exposure only; allethrin and vitamin C; allethrin and degumming of silkworm cocoons at doses of 100 mg/kg, 200 mg/kg, and 300 mg/kg. This study used 25 male rats (Mus musculus L.). The observations were conducted at the morphology of the testes including the weight, the colour, a the consistency, and the histology of the testes including the diameter of the seminiferous tubules. It was also examined the thickness of the germinal epithelium of the seminiferous tubules, the number of Leydig cells, the number of spermatogonia, spermatocytes, and spermatids. The results showed that degumming of the silkworm cocoons could improve the colour and the consistency. It increased the testicular weight, the diameter of seminiferous tubules, the thickness of germinal epithelium of seminiferous tubules, the number of Leydig cells, the number of spermatogonia, the spermatocytes and spermatids as well. The silkworm cocoon degumming at a dose of 300 mg/kgBW was the best dose in improving the morphological and histological features of the testes of allethrin-exposed male mice (Mus musculus L.).en_US
