    Abstract
    Effect of 2,4-Dichlorophenoxyacetate (2,4- D) Application at Several Media Concentrations on Black Orchid (Coelogyne pandurata) In Vitro Culture, supervised by Emmy Harso Kardhinata. This study aims to determine the effect of MS media concentration and 2,4-D and their interaction on the growth of black orchid in vitro. This research was conducted at the Tissue Culture Laboratory, Alifa Agricultural Research Center (ALIFA-ARC), from April to June 2023. This study used a Completely Randomized Design (CRD) method with 2 treatment factors. The first factor is the concentration of MS Media: half MS and full MS. The second factor is the concentration of 2,4 D: (0 mg/L, 2 mg/L, 4 mg/L, 6 mg/L and 8 mg/L). There were 10 treatment combinations with 3 replications resulting in 50 experimental units. The parameters measured were: percentage of living explants (%), percentage of contaminated explants (%), number of shoots per explant (fruit), shoot height (cm), number of roots (fruit) and root length (cm). The results showed that the treatment of MS media concentration had no significant effect on black orchid tissue culture on all observed parameters. The treatment of 2,4 D concentration had no significant effect on black orchid tissue culture on all observed parameters. The interaction between MS media concentration and 2,4 D had no significant effect on black orchid tissue culture on all observed parameters. In addition, it can be seen that M0D1 treatment is the best combination for all observed parameters.
