dc.description.abstractUsing a Combination of ZPT Picloram and Kinetin, supervised by Revandi Iskandar Muda Damanik. This research aims to analyse the content of secondary metabolites in the form of flavanoid and tannin compounds in black cumin plant callus using endogenous chemical elicitors (sucrose) on MS liquid media with a combination of Growth Regulators Picloram and Kinetin. This research was conducted at the Tissue Culture Laboratory of the Faculty of Agriculture and the Phytochemistry Laboratory of the Faculty of Pharmacy, University of North Sumatra from February 2023 to July 2023. This study used the Complete Randomised Design (CRD) method with 2 treatment factors, 12 combinations and each treatment had 3 repetitions. The first repetition factor is the concentration of Growth Regulators on MS media (M): M1 (Picloram 1 ppm and Kinetin 1 ppm), M2 (Picloram 1 ppm and Kinetin 2 ppm), M3 (Picloram 2 ppm and Kinetin 1 ppm), and M4 (Picloram 2 ppm and Kinetin 2 ppm). The second factor was sucrose concentration in liquid MS media: S1 (25 g/l MS media), S2 (30 g/l MS media), and S3 (35 g/l MS media). Parameters observed were callus morphology, growth index, SCV (Settled Cell Volume), PCV (Peacked Cell Volume) and secondary metabolite content analysis. The results showed that the provision of sucrose concentration in liquid MS media and the concentration of ZPT picloram and kinetin did not significantly affect all parameters.en_US
